By selecting appropriate materials and methods, microneedle-based vaccines with strong mechanical properties and variable forms can be fabricated for monitoring or augmenting human immunity. The mechanism of skin immunity, classification and fabrication approaches of microneedles, as well as their applications in sampling and therapy, are discussed.

Summary: A novel peptide called Decapeptide-12 was modified by adding a palmitate chain to enhance its transcutaneous penetration, and the modified palm-peptide showed improved skin retention due to increased lipophilicity. The combinatory approach of molecular modification, chemical penetration enhancers, and microneedle patch proved to be effective in enhancing skin permeation of the peptides, potentially offering a promising solution for treating hyperpigmentation without the adverse effects associated with traditional treatments like hydroquinone.

Summary: This study investigated the feasibility of using minimally invasive physical ablative techniques for delivering naloxone via the skin. These techniques reduced the lag time to naloxone delivery and significantly increased the amount of naloxone delivered in 1 h and over 24 h compared to passive delivery. Sonophoresis and laser showed the highest delivery in 1 h, followed by dermaplaning. However, there were inconsistencies in the in vitro permeation profile of naloxone with the ultrasound frequency used. With proper optimization, these techniques can potentially be applied for systemic naloxone delivery in opioid emergencies and disaccustoming purposes.

Summary: A latch applicator, consisting of simple plastic parts and operated by thumb force, was developed for efficient and cost-effective administration of dissolving microneedles (DMN). The optimized latch applicator demonstrated successful insertion of the drug-loaded DMN into the skin and showed higher antibody production compared to intramuscular injection in an immunization test. This latch applicator has the potential to greatly improve drug delivery, including vaccines, with its effective DMN insertion and competitive price.

Summary: A novel photo-thermally dissolvable hyaluronic acid microneedle patch with biologics encapsulation has been developed for the treatment of psoriasis. The microneedles possess suitable mechanical strength and biocompatibility, allowing for minimal immune response upon skin puncture. The rapid melting and release of encapsulated IL-17 mAbs under near-infrared light irradiation provide an effective anti-inflammatory effect around psoriasis lesions. This multifunctional microneedle patch effectively reduces psoriasis-like skin inflammation in an animal model and offers a safer and more effective strategy for psoriasis treatment and other chronic inflammatory autoimmune diseases.

Summary: This study aimed to develop polymeric dressings, microfibers, and microneedles loaded with ceftriaxone to treat diabetic wounds and promote faster healing. The formulations were optimized and subjected to physicochemical tests. The results showed that the developed dressings, microfibers, and microneedles exhibited favorable bioadhesion, tear strength, erythema, hydration, pH, and drug release properties. In vitro studies demonstrated promising flux, permeation coefficient, and time lag values in wounded skin, and in vivo tests in diabetic rats showed healing time below 14 days. These formulations have great potential for addressing the challenges of chronic wounds, such as diabetic foot, and improving clinical outcomes.

Summary: This study evaluates the performance of Laponite in lecithin/gelatin composites and maltodextrin/sodium ascorbate mixtures. Nanoparticles were prepared using the gelatin desolvation method and turned into films via the solvent-casting method. The optimal composites were developed with 8.8 mg of Laponite, reducing particle size and avoiding agglomeration. Drug release in LGL MAS was fitted to first-order and Korsmeyer-Peppas models.
