Summary: Pain after cancer is often underestimated and undertreated. Precision medicine offers a way to classify patients into different subgroups based on their individual characteristics and tailor treatment accordingly. The application of precision medicine to post-cancer pain is an emerging issue, particularly in identifying and treating nociplastic pain. The 2021 IASP clinical criteria provide a framework for classifying and treating nociplastic pain in cancer patients, and the CANPPHE Network aims to implement and improve this approach through a diagnostic manual and case illustrations.

Molecular imaging of liver inflammation using an anti-VCAM-1 nanobody

Summary: In this study, researchers found that molecular imaging of VCAM-1 could be a non-invasive alternative to biopsy for detecting liver inflammation in chronic liver disease. By using radiolabeled anti-VCAM-1 nanobody and SPECT, they were able to quantify liver uptake and correlate it with histological inflammatory scores and molecular markers. The diagnostic power of this imaging tool to detect liver inflammation was excellent, suggesting its potential for clinical use.

Summary: mAbs have limitations in their distribution and penetration in tumor microenvironment, as well as their ability to reach the brain. Nanobodies, being smaller in size, possess superior abilities in tumor penetration and infiltration into brain tumors. However, the rapid clearance of nanobodies from circulation may affect their suitability for therapy, although their noncovalent binding to albumin can help increase their serum half-life.

This overview provides insights into nanobody-based diagnostics and therapeutics that have been tested in clinical trials and highlights upcoming preclinical developments.

Summary: Macrophages play a vital role in the body, and anti-inflammatory macrophages targeting CD206 are involved in various diseases. This study evaluated the safety and biodistribution of a PET tracer targeting CD206 in human subjects, and found that the tracer was safe, rapidly cleared from the blood, and enabled high contrast imaging at 90 minutes after injection. Preliminary results showed higher tumor uptake in patients with disease progression.

Optimizing sexuality of young women with breast cancer: how can the breast clinic help?

Summary: This study explores how specialized breast clinics can help young women with their sexual health questions or problems by fulfilling their information and social support needs. The study finds that young women lack information on sexual issues and the information provided is not sufficiently tailored to them. The empathy and communication skills of healthcare providers play an important role in whether sexual issues can be discussed. Finally, the study suggests that more attention should be paid to the partner's well-being and the relationship.

Summary: Blockade of the PD-1/PD-L1 immune checkpoint axis alleviates the inhibition of tumor-infiltrating lymphoid cells but weakly induces their expansion. Exogenous cytokines can further expand lymphoid cells and synergize with alpha PD-L1 therapy. Local delivery of cytokines and alpha PD-L1 therapeutics to immune cell-containing melanoma tumors effectively mediates anti-tumor activity and sensitizes the tumor microenvironment mainly through impacting NK cells.

Nanobody-based pannexin1 channel inhibitors reduce inflammation in acute liver injury

Summary: This study successfully developed nanobody-based inhibitors targeting Pannexin1 channels, which showed affinity for both murine and human Pannexin1 proteins. These nanobodies were able to block the release of adenosine triphosphate and display anti-inflammatory effects in vitro, as well as in a relevant mouse model of acute liver disease. The findings suggest that these nanobodies hold promise as therapeutic agents for inflammatory disorders and provide novel tools for further research on the role of Pannexin1 channels in physiological and pathological processes.

Development and evaluation of nanobody tracers for noninvasive nuclear imaging of the immune-checkpoint TIGIT

Summary: This study generated a panel of nanobodies targeting human or mouse TIGIT, and assessed their performance in vitro and in vivo. The nanobodies demonstrated excellent affinity and specific binding to TIGIT, and showed promising diagnostic potential for noninvasively imaging high TIGIT expression within the tumor. These findings hold promise for improving patient stratification and therapy response.

Safe implementation of robotic right colectomy with intracorporeal anastomosis

Summary: This study demonstrates that robotic right colectomy with intracorporeal anastomosis can be safely implemented without increasing morbidity, although it requires a learning phase with longer procedure times compared to laparoscopy. The conversion rate from laparoscopy to robotic surgery is significantly reduced.
