Ezrin Inhibition Overcomes Acquired Resistance to Vemurafenib in BRAFV600E-Mutated Colon Cancer and Melanoma Cells In Vitro

Summary: This study identified ezrin protein as a key factor in acquired resistance to vemurafenib in BRAFV600E-mutated colon cancer and melanoma cells. These findings support further exploration of combined BRAF inhibitors and actin-targeting drugs as a potential therapeutic approach in BRAFV600E-mutated cancers.

Sustained release hydrogel for durable locoregional chemoimmunotherapy for BRAF-mutated melanoma

Summary: The wide prevalence of BRAF mutations in diagnosed melanomas has driven the development of BRAF inhibitors in combination with immune checkpoint blockade for the treatment of advanced disease. This study demonstrates the benefits of a novel thermosensitive hydrogel for the co-delivery of a small molecule BRAF inhibitor with immune checkpoint blockade antibody, improving the therapeutic index of the combination therapy.

Through clinical trials, it was found that the maximum tolerated dose of Palbociclib in treating these patients was 25mg, while also achieving some clinical responses.

Summary: The outcomes of patients with metastatic melanoma discontinuing BRAF-targeted therapy for cumulative toxicity after sustained response are unknown. This study analyzed patients with sustained responses (median treatment duration 59.4 months), finding 12- and 24-month progression-free survival rates of 70.8% and 58.3%, respectively. Complete response and negative circulating tumor DNA at time of discontinuation are promising prognostic biomarkers in this setting.

Recent advances in molecular targeted therapy for unresectable and metastatic BRAF-mutated melanoma

Summary: The clinical outcome of BRAF-mutated advanced melanoma has been improved by both molecular targeted therapies and immune checkpoint inhibitors. Long-term follow-up data reveal durable clinical responses in patients receiving first-line combinations of BRAF inhibitors plus MEK inhibitors, particularly those showing a complete response. Clinical outcomes are also associated with the lactate dehydrogenase levels and the number of metastatic organs.

Vitamin D Modulates the Response of Patient-Derived Metastatic Melanoma Cells to Anticancer Drugs

Summary: Preconditioning of patient-derived melanoma cells with 1,25(OH)(2)D-3 enhances the inhibitory effect of cediranib and vemurafenib and reduces mitochondrial respiration parameters. Moreover, cancer-associated fibroblasts play a crucial role in melanoma growth. Thus, 1,25(OH)(2)D-3 could be considered as an adjuvant agent in the treatment of malignant melanoma.

Panniculitis does not predict clinical response in patients with advanced melanoma under targeted therapy

Summary: In this study, the association between panniculitis occurrence during targeted therapy and the outcome of metastatic melanoma was explored. The results showed no significant association between panniculitis and clinical response to targeted therapy. According to the literature review, panniculitis mainly affects young women during targeted therapy, with variable delay to onset, and usually only affects the lower limbs or is associated with other clinical signs.

The use of minimal residual disease in thoracic oncology: Gaps between promises and the on-the-ground reality of daily practice

Summary: The assessment of minimal residual disease (MRD) from blood samples has potential in optimizing patient care for resected non-small cell lung carcinoma (NSCLC). Evaluating MRD status can improve overall survival for early stage NSCLC patients and reduce the toxic effects and financial burden of treatment. Clinical trials have examined MRD in early stage NSCLC, highlighting the need to bridge the gap between research and routine practice. Further action is necessary to assess the relevance of MRD detection in interventional clinical studies, comparing different techniques, time points, and cutoffs. This article investigates and provides recommendations for optimizing MRD evaluation in NSCLC.

Summary: As new SARS-CoV-2 variants emerge, it is important to improve the efficiency and availability of viral genome sequencing, especially in samples with low viral load. In this study, retrospective SARS-CoV-2 genome sequencing was performed on 175 positive samples, and the analysis successfully detected Delta and Omicron variants in low viral load samples. The results showed that there was no significant difference in viral load between patients infected with the Omicron variant and those infected with the Delta variant.

Hidradenitis suppurativa and social disadvantage: A nationwide administrative data study of the French hospitalized population

RNA NGS can be a viable alternative to replace ROS1 IHC as a screening method.

Quantitative multiplexed imaging technologies for single-cell analysis to assess predictive markers for immunotherapy in thoracic immuno-oncology: promises and challenges

Summary: Over the past decade, there has been a revolution in cancer treatment with the shift towards targeted molecular therapies and immune-based therapies, particularly immune-checkpoint inhibitors (ICIs). However, not all patients with advanced non-small cell lung cancer (aNSCLC) benefit equally from immunotherapy, and there is a need for additional predictive markers for ICIs efficacy. Multiplex immunostaining, which evaluates multiple molecules in the tumor at the same time, shows promise in optimizing patient selection for ICIs. This review aims to explore the role of multiplex immunostaining in immuno-thoracic oncology and its current advantages and limitations in clinical practice.
