Summary: The standard frontline treatment for metastatic colorectal cancer (CRC) is cytotoxic chemotherapy combined with a biologic agent. Predictive biomarkers such as mismatch repair (MMR) status and RAS and BRAF mutation status play an important role in treatment selection. Other factors considered include primary tumor location, intent of therapy, potential toxicity, patient age, comorbidities, and patient preference. Future studies may provide further personalized therapy options based on additional predictive biomarkers for this heterogeneous disease.

Summary: This study assessed the cost-effectiveness of cetuximab in combination with chemotherapy for first-line treatment of KRAS wild type mCRC in Indonesia. The results showed that cetuximab plus chemotherapy did not significantly improve life years gained compared to chemotherapy alone. The cost-effectiveness ratio was high, indicating that it is not cost-effective.

Wild-Type KRAS Is Required for Panitumumab Efficacy in Patients With Metastatic Colorectal Cancer

Summary: The purpose of this study is to determine the impact of KRAS gene mutations on the efficacy of panitumumab monotherapy for metastatic colorectal cancer (mCRC). Through testing on 463 patients, it was found that 43% of patients had KRAS mutations. The results showed that panitumumab significantly improved overall survival and progression-free survival for patients with wild-type KRAS, while no significant efficacy was observed for patients with KRAS mutations. Therefore, KRAS status should be taken into account when selecting mCRC patients for panitumumab monotherapy.

Summary: High baseline plasma AREG levels in CRC patients are associated with shorter PFS with cetuximab treatment, but not with PFS or overall survival after chemotherapy failure. AREG may decrease the anti-proliferative effect of cetuximab, but loses this effect after acquiring resistance. Plasma AREG could potentially serve as a biomarker for predicting clinical outcomes with cetuximab-based chemotherapy.

Summary: This study investigated the impact of metastatic patterns on prognosis and prediction of maintenance therapy in patients with colorectal cancer. The results showed that multiple organ metastases were associated with poorer survival outcomes, and patients with liver metastasis plus >= two additional sites had the worst prognosis. In patients with multiple organ metastases, maintenance therapy with panitumumab showed good efficacy.

EGFR-amplified tumors are usually MSS and HER2 nonamplified, with a higher median fraction of genome altered. Patients with EGFR amplification tend to have longer overall survival and better outcomes when exposed to anti-EGFR-based therapy.

Summary: Myxopapillary ependymoma (MPE) is a primary CNS tumor characterized by its indolent malignancy involving the spinal conus medullaris, Filum terminale or cauda equina. We report a rare case of MPE recurring in the pelvic soft tissue and metastasizing to the pleura and lungs. Despite resistance to multiple treatments, including surgery, radiotherapy, chemotherapy, and temozolomide, the combination regimen of olaparib-temozolomide (OT) targeting somatic BRCA1 mutations has achieved stable disease after 10 cycles.

Robotic versus freehand CT-guided radiofrequency ablation of pulmonary metastases: a comparative cohort study

Summary: The purpose of this study was to evaluate the feasibility, safety, and accuracy of robotic radiofrequency ablation (RFA) for pulmonary metastases compared to conventional freehand placement. The study found that robotic RFA was technically successful with high accuracy and fewer manipulations, leading to improved safety and reduced complications compared to freehand placement.

Summary: This study identified miR-148a-3p as a potential biomarker for trastuzumab-based therapy in HER2-positive oesophago-gastric cancer patients. The results showed that the expression level of miR-148a-3p is associated with patient survival and progression rates.
