A novel human volunteer pain model using contact heat evoked potentials (CHEP) following topical skin application of transient receptor potential agonists capsaicin, menthol and cinnamaldehyde

The discovery of transient receptor potential (TRP) receptors has advanced understanding of temperature sensation, and pre-clinical studies have identified TRP as major novel analgesic targets in inflammatory and neuropathic pain states. We systematically investigated the sensory effects and interactions of TRP agonists capsaicin (TRPV1), menthol (TRPM8) and cinnamaldehyde (TRPA1) applied topically to the skin in 14 healthy human participants. Capsaicin lowered heat pain thresholds while warm detection thresholds were unchanged, suggesting an effect purely on nociceptor nerve fibres. The amplitude of contact heat-evoked potentials (CHEP) and evoked pain ratings were negatively correlated after capsaicin, whereas CHEP had been correlated positively without capsaicin in a previous volunteer study. Menthol caused cold hypersensitivity and cinnamaldehyde caused heat hypersensitivity, but neither had an effect on evoked potentials. The CHEP after application of capsaicin show features observed in some patients with painful neuropathy, and could provide a model for development of novel analgesics, particularly TRPV1 antagonists. Global importance analysis: An interpretability method to quantify importance of genomic features in deep neural networks

Dose-Related Inhibition of Capsaicin Responses by Cannabinoids CBG, CBD, THC and their Combination in Cultured Sensory Neurons

Summary: CBG, CBD, and THC individually demonstrated potent dose-related inhibition of capsaicin responses in DRG neurons in vitro, and their combination showed enhanced effectiveness. The high correlation between cannabinoid-mediated calcium influx and reduction of capsaicin responses suggests potential synergistic effects of CBG, CBD, and THC when applied together. Further studies are needed to explore the efficacy and tolerability of these combinations in vivo.

Framed Building Response to Tunnelling on Different types of Foundations

Summary: The objective of this study is to investigate and compare the effect of tunnelling on the response of a framed building on different types of foundations. The ovalling effect of a circular tunnel on the displacement of the building was analyzed using FEM-based software ABAQUS. The study considered three different types of foundations: isolated foundation, mat foundation, and pile foundation. The results showed that the maximum displacement of the building frame occurred when the tunnel was situated just below the center axis of the building frame for all types of foundations. Building constructed over isolated foundation showed the highest displacement compared to building constructed over mat foundation and pile foundation for all locations and sizes of tunnel.

Study of Tunnelling-Induced Response of Building Frame with Shake Table Test

Summary: This paper investigates the effect of tunnelling on the response of a framed building on different types of foundations through shake table test. The results show that the ovalling effect of circular tunnel mainly affects the displacement of the building, and its influence diminishes as the distance between the building frame and tunnel increases. Furthermore, the type of foundation used for the building also has varying degrees of influence on the effect of the tunnel.

Nerve and Vascular Biomarkers in Skin Biopsies Differentiate Painful From Painless Peripheral Neuropathy in Type 2 Diabetes

The ratio of CGRP:vWF in sub-epidermal nerve fibers showed a significant decrease in painful-DPN compared to painless-DPN, suggesting that increased dermal vasculature may differentiate painful-DPN from painless-DPN.

Evaluation of forward head posture and thoracic kyphosis in migraine
