Summary: Cell-type specific DNA methylation can be used to accurately count different leukocyte subsets in blood. In this study, the authors developed targeted DNAm assays and used digital droplet PCR to measure DNAm levels in venous blood and dried blood samples. The results showed good correlations between epigenetic estimates and conventional cell counting methods, and dried blood samples facilitated self-sampling for easier testing accessibility.

Summary: The study aimed to evaluate a procedure in which urine culture was performed based on fixed cut-off values of urine sediment analysis to prevent unnecessary negative urine cultures. Urine samples were analyzed from January 2018 to August 2018 in the urology outpatient department. Results showed that adjusting the cut-off values of sediment analysis could significantly reduce the number of urine cultures and unnecessary costs.

Summary: P2RX7 is a poorly selective ATP-gated ion channel with important physiological and pathological implications, making it a potential therapeutic target in several diseases. Novel screening methods based on flow cytometry and spectral flow cytometry have been developed to efficiently measure P2RX7 activity and assess the efficacy of modulators. Spectral cytometry appears to be a useful tool for drug candidate screening by simultaneously evaluating multiple biological activities and correcting for the autofluorescence of compounds screened.

Summary: This study evaluated the diagnostic potential of an anti-TRBC1 monoclonal antibody for the identification of T-cell non-Hodgkin lymphomas (NHL). Flow cytometry analysis showed that TRBC1 expression provides a robust method for T-cell clonality assessment with high sensitivity and good correlation with complementary methods. The findings suggest that TRBC1 can be integrated into routine lymphoma screening strategies.

Summary: We propose a theory for high-throughput genetic interaction screens, which extends the Multiplexing using Spectral Imaging and Combinatorics (MuSIC) approach to create 100,000 spectrally unique MuSIC barcodes. Experimental testing of this theory could revolutionize genetic perturbation technology and knowledge of genetic function. Additionally, a genome-scale spectral barcode library could have widespread applications in genetic screening and lineage tracing.

In vitro micropropagation techniques, such as tissue culture, offer a promising tool for cost-effective and efficient propagation. This study developed a rapid and efficient in vitro multiplication protocol for Maerua crassifolia and demonstrated successful acclimatization and genetic fidelity of the micropropagated plants.

Summary: The Sysmex UF-4000i fluorescence flow cytometry analyzer can be used as an alternative method to urine cultures to rule out negative urinary tract infections. Bacteria count at a cut-off of 100/μL and/or leukocytes count ≥ 45/μL were found to be the optimal indicators for positive culture results.
