Mysterious link between iron overload and CDKN2A/2B

Persistent oxidative stress has been associated with carcinogenesis. Iron overload is considered one such condition that causes oxidative stress. Epidemiological studies support a close link between iron overload and carcinogenesis. Reportedly, regular semiannual phlebotomies reduced cancer risk in an otherwise normal population. More specifically, genetic hemochromatosis, chronic viral hepatitis, ovarian endometriosis and asbestosis induce iron overload, which can lead to hepatocellular carcinoma, ovarian carcinoma or mesothelioma in humans. Through a combination of animal experiments and microarray analyses, homozygous deletion of CDKN2A/2B has been recognized as one of the major target genes involved in iron overload-induced carcinogenesis. CDKN2A/2B are the second most frequently inactivated tumor suppressing genes in human cancers. Currently, when infection is becoming sufficiently controlled worldwide, iron regulation may be the next target for human longevity.

Summary: The study utilized electron paramagnetic resonance spectroscopy to quantify different reactive oxygen species generated by NTP in the presence of antioxidants, demonstrating significant scavenging effects of antioxidants on reactive oxygen species. The importance of redox cycling of Asc/DHA and metabolites of DHA, as well as the impact of ROS-reducing compounds like EGCG, were highlighted in the study for NTP application in cells and tissues. Further research is needed to explore the interaction between ROS and biomolecules to enhance the medical applications of NTP.

Non-thermal plasma-induced DMPO-OH yields hydrogen peroxide

Summary: Recent developments in electronics have enabled the medical applications of non-thermal plasma (NTP), which elicits reactive oxygen species (ROS) and reactive nitrogen species (RNS), such as hydroxyl radical ((OH)-O-center dot), hydrogen peroxide (H2O2), singlet oxygen (O-1(2)), superoxide (O-2(center dot-)), ozone, and nitric oxide at near-physiological temperatures. In preclinical studies or human clinical trials, NTP promotes blood coagulation, eradication of bacterial, viral and biofilm-related infections, wound healing, and cancer cell death. Studies using electron paramagnetic resonance (EPR) spectroscopy have shown that NTP-induced reactive oxygen species (ROS) can be effectively scavenged in the presence of biocompatible reducing agents.

Summary: Poly (rC)-binding proteins 1 and 2 (PCBP1/2) play an important role in mesothelial cells and malignant mesothelioma (MM) by conferring resistance to ferroptosis. Knockdown of PCBP2 decreases the expression of transferrin receptor 1 (TfR1) and ferritin heavy chain (FTH), inhibits cell proliferation, and increases sensitivity to ferroptosis in MM cells.

Ferroptosis-dependent extracellular vesicles from macrophage contribute to asbestos-induced mesothelial carcinogenesis through loading ferritin
