Quantitation of paclitaxel and its two major metabolites using a liquid chromatography-electrospray ionization tandem mass spectrometry

A sensitive and selective liquid chromatographic-tandem mass spectrometric (LC-MS/MS) method for the determination of paclitaxel (Taxol) and its two major metabolites in human plasma has been developed. Samples were prepared after liquid-liquid extraction and analyzed on a C(18) column interfaced with a Q-Trap tandem mass spectrometer. Positive electrospray ionization was employed as the ionization source. The mobile phase consisted of acetonitrile-water (0.05% formic acid) (65:35) at the flow rate of 0.25 mL/min. The analytes and internal standard docetaxel were both detected by use of multiple reaction monitoring mode. The method was linear in the concentration range of 0.5-500.0 ng/mL for paclitaxel, 6 alpha-hydroxypaclitaxel and p-3'-hydroxypaclitaxel, respectively. The lower limit of quantification (LLOQ) was 0.5 ng/mL for paclitaxel, 6 alpha-hydroxypaclitaxel and p-3'-hydroxypaclitaxel, respectively. The intra- and inter-day relative standard deviation across three validation runs over the entire concentration range was less than 8.18%. The accuracy determined at three concentrations was within +/-10.8% in terms of relative error. The total run time was 7.0 min. This assay offers advantages in terms of expediency, and suitability for the analysis of paclitaxel and its metabolites in various biological fluids. (C) 2011 Published by Elsevier B.V.

(±)-Atrachinenins A-C, Three Pairs of Caged C27 Meroterpenoids from the Rhizomes of Atractylodes chinensis

Structural elucidation was done through various spectroscopic techniques and a plausible biosynthetic pathway was proposed. In situ Chemical Profiling and Imaging of Cultured and Natural Cordyceps sinensis by TOF-SIMS

Summary: Time-of-flight secondary ion mass spectrometry (TOF-SIMS) is a sensitive surface analytical technology that allows for simultaneous acquisition of diverse chemical components and their precise locations on the surfaces of samples. In this study, a new sample preparation strategy was developed to perform in situ chemical analysis of Cordyceps sinensis using TOF-SIMS. The technique was successfully applied to compare natural and cultured Cordyceps sinensis, revealing important differences in their chemical distributions. Overall, TOF-SIMS is considered an advanced and promising analytical technology for the research of traditional Chinese medicines.

Oriented Layered Graphene Oxide Pad Favoring High Loading Capacity and Stability for High-Throughput Chemical Screening

Summary: Carbon nanomaterials, particularly graphene oxide (GO), have great potential in materials science and biomedicine. However, GO lamelayers tend to aggregate and are difficult to disperse in solution, resulting in low loading efficiency. To address this issue, oriented layered graphene oxide pad (OGO) was synthesized as a carrier to improve loading capacity and stability. The OGO structure showed good reproducibility and recyclability, with open channels and low pore curvature for better flow and higher fishing sites density. The CMP@OGO achieved 1.5 to 2 times higher loading capacity compared to GO. Moreover, the established CMP@OGO exhibited stability and high reuse rate, making it a promising platform for active compound screening.
