Summary: A simple, sensitive, and rapid LC-MS/MS method was developed for the simultaneous determination of BIC, M7, and M8 active metabolites in SD rat plasma. The method showed excellent linearity and accuracy for quantitation in plasma samples. Pharmacokinetic analysis revealed improved parameters in the BIC-NPs group compared to the BIC group, with a significant increase in AUC(0-t) for BIC and a double peak phenomenon observed for M7.

Summary: Thiopurines are commonly used in the treatment of chronic autoimmune conditions and leukemia, but may cause gastrointestinal toxicity. This study investigated the metabolite concentrations of thiopurines in intestinal cells, finding associations between metabolite concentrations and cytotoxicity. The findings suggest that short growth times before treatment should be used to study the association between thiopurine metabolite concentrations and cytotoxicity activity in vitro.

Summary: Naringin is a dietary flavonoid glycoside with multiple bioactivities. The results revealed the visible individual difference and low urinary recovery of flavonoid metabolites in the excretion of naringin, which may be helpful for further study to understand the in vivo behavior and action mechanism of naringin.

Summary: The aim of this study was to determine the excretion of methylone and its metabolites in sweat after controlled doses of 50, 100, 150, and 200 mg of methylone were ingested by twelve healthy volunteers in a clinical trial. Methylone and its metabolites, 4-hydroxy-3-methoxy-N-methylcathinone (HMMC) and 3,4-methylenedioxycathinone (MDC), were detected in sweat patches using liquid chromatography-tandem mass spectrometry. Methylone and MDC were found in sweat after 2 hours and reached their highest concentration at 24 hours following the administration of doses. However, HMMC was not detectable at any time interval. Sweat has proven to be a suitable matrix for determining the concentration of methylone and its metabolites in clinical and toxicological studies, indicating recent drug consumption.

Summary: Atrachinenins A-C (1-3), isolated from the rhizomes of Atractylodes chinensis, are meroterpenoids with novel C-27 skeletons. Atrachinenin A (1) features a unique cage-like skeleton, while atrachinenin B (2) has an additional peroxyl bridge ring, forming a complex cyclic ring system. Structural elucidation was done through various spectroscopic techniques and a plausible biosynthetic pathway was proposed. Atrachinenin A exhibited potential TGF-beta inhibitory activity at 25 μmol/L.

Summary: Time-of-flight secondary ion mass spectrometry (TOF-SIMS) is a sensitive surface analytical technology that allows for simultaneous acquisition of diverse chemical components and their precise locations on the surfaces of samples. In this study, a new sample preparation strategy was developed to perform in situ chemical analysis of Cordyceps sinensis using TOF-SIMS. The technique was successfully applied to compare natural and cultured Cordyceps sinensis, revealing important differences in their chemical distributions. Overall, TOF-SIMS is considered an advanced and promising analytical technology for the research of traditional Chinese medicines.

Summary: Carbon nanomaterials, particularly graphene oxide (GO), have great potential in materials science and biomedicine. However, GO lamelayers tend to aggregate and are difficult to disperse in solution, resulting in low loading efficiency. To address this issue, oriented layered graphene oxide pad (OGO) was synthesized as a carrier to improve loading capacity and stability. The OGO structure showed good reproducibility and recyclability, with open channels and low pore curvature for better flow and higher fishing sites density. The CMP@OGO achieved 1.5 to 2 times higher loading capacity compared to GO. Moreover, the established CMP@OGO exhibited stability and high reuse rate, making it a promising platform for active compound screening.

Summary: An efficient approach for the synthesis of 1,2-diaryl diketones was developed from readily available alpha-methylene ketones by catalysis of I-2. In the same oxidation system, a novel one-pot procedure was established for the construction of antiviral and anticancer quinoxalines, showing good functional group tolerance with moderate to excellent yields. The synthesized compounds exhibited inhibitory effects on viral entry and cytotoxicity against cancer cells.
