An improved HPLC-UV method for the simultaneous quantification of triterpenic glycosides and aglycones in leaves of Centella asiatica (L.) Urb (APIACEAE)

The simultaneous quantification of madecassoside, asiaticoside, madecassic acid and asiatic acid in Centella asiatica by HPLC-UV is proposed. Asiaticoside was used as reference for the quantification of heterosides and asiatic acid for aglycones. The evaluation of the extraction efficiency of the four molecules led to use Soxhlet extraction for 8 h. The method was validated and was found to be accurate in the concentration range of 1.0-3.0 mg/ml for asiaticoside and 0.5-2.0 mg/ml for asiatic acid with CV <3% for all investigated compounds. LOD and LOQ were, respectively, 0.0113 and 1.0 mg/ml for asiaticoside and 0.0023 and 0.5 mg/ml for asiatic acid. This method was shown to be convenient for routine analysis of samples of C. asiatica. A Validated HPLC-PDA-HRMS Method to Investigate the Biological Stability and Metabolism of Antiparasitic Triterpenic Esters

Summary: Pentacyclic triterpenes (PTs) are commonly found in medicinal plants with antiparasitic effects. This study aimed to determine the stability of triterpenic esters in different biological-like media and their main microsomal degradation products. The results showed that these esters exhibited good stability in aqueous and plasma solutions, but degradation was observed in acidic media. Moreover, hepatic microsomal metabolism varied depending on the concentration and position of the ester link, suggesting different metabolic pathways for these compounds.

5'AMP-activated protein kinase: an emerging target of phytochemicals to treat chronic inflammatory diseases

Summary: Inflammation is a defensive response of the organism to various injuries. Chronic inflammatory diseases are challenging to treat, and phytochemicals may provide an interesting alternative approach. AMP-activated protein kinase plays an important role in regulating inflammatory responses. This review summarizes studies on phytochemicals that activate AMPK to improve chronic inflammatory disease models, and highlights factors to consider when investigating AMPK in the context of phytochemicals.

Bioactive Clerodane Diterpenoids from the Leaves of Casearia coriacea Vent

Summary: Casearia coriacea Vent., a plant endemic to the Mascarene Islands, exhibited potent antiplasmodial activity through the presence of three identified compounds, caseamembrin T (1), corybulosin I (2), and isocaseamembrin E (3), with IC50 values ranging from 0.25 to 0.51 μg/mL. While these compounds also showed antileishmanial and antitrypanosomal properties, they exhibited selectivity towards the malaria parasite with selectivity indices ranging from 6 to 12 and 25 to 100, respectively. Additionally, compounds 1-3 demonstrated promising activity against pancreatic carcinoma cells (IC50 < 1 μg/mL). However, the main compound, caseamembrin T (1), displayed significant toxicity when evaluated on zebrafish embryos.

Summary: By performing LC-MS/MS analysis and molecular networking, the main compounds in Terminalia leiocarpa were identified as ellagic acid and derivatives, gallic acid, astragalin, rutin, quinic acid, and fructose. Among them, ellagic acid and gallic acid showed good anthelmintic activity against C. elegans. Other unidentified compounds may also contribute to the observed activity.
