A fast method for the quantification of methylamine in fermentation broths by gas chromatography

The objective of this study was to develop a method for the quantitative analysis of the methylamine concentration in fermentation broths of Hyphomicrobium zavarzinii ZV 580 cultures. For this purpose an established method for the quantification of free amino acids in such matrices was adapted and validated. The detection limit was 10 mu M, the calibration curve showed good linearity (R-2 = 0.9998) in the concentration range between 0.1 and 8 mM. The standard deviation of the injection-to-injection reproducibility (n = 10) of the retention coefficient was <1%, that of the peak area <5%. In case of the sample-to-sample reproducibility (n = 8), the standard deviation was <5% for the retention coefficient and <10% for the peak area. The validated method was successfully applied for monitoring a fed-batch bioprocess (starting volume: 8 L, initial methylamine hydrochloride concentration: 10 mM) producing a dye-linked formaldehyde dehydrogenase in H. zavarzinii ZV 580. Generation of Recombinant Primary Human B Lymphocytes Using Non-Viral Vectors

Isolation of primary human B lymphocytes from tonsils compared to blood as alternative source for ex vivo application

Noxic effects of polystyrene microparticles on murine macrophages and epithelial cells

Summary: The study found that macrophages can effectively ingest microplastic particles, while epithelial cells did not show significant ingestion of microplastics. Summary: The intestinal cells of the earthworm Eisenia fetida are suitable for studying the toxic effects of environmental pollutants on a cellular level. Exposure to pollutants and soil elutriates led to a decrease in metabolic activity of the cells. Additionally, microplastic particles did not show active uptake, but had dose and size dependent effects on metabolism that may correlate with organismic effects.

Microplastic Contamination of Composts and Liquid Fertilizers from Municipal Biowaste Treatment Plants: Effects of the Operating Conditions

Summary: High-caloric-value household biowaste is a desirable substrate for biogas and fertilizer production, but it is often contaminated by plastic packaging. This study found that fertilizer from biowaste treatment plants had significant plastic contamination, while fertilizer from agricultural biogas plants and greenery composts had minimal contamination. The level of contamination appeared to be influenced by operating conditions, with shredding of incoming material increasing the degree of contamination.

Influence of NH3 and NH4+on anaerobic digestion and microbial population structure at increasing total ammonia nitrogen concentrations

Summary: Despite extensive research, the effects of NH3 and NH4+ on anaerobic digestion and associated microorganisms are still not fully understood. This study investigated the effects of different NH3/NH4+ ratios on microorganisms involved in anaerobic digestion of two substrates. It was found that bacteria, particularly gram-positive bacteria, were affected by total ammonia nitrogen (TAN), while archaea were largely influenced by NH3 concentration. Adjusting the NH3/NH4+ ratio can avoid inhibition and stabilize biogas production in nitrogen-rich substrates.

Role of Residual Monomers in the Manifestation of (Cyto)toxicity by Polystyrene Microplastic Model Particles

Summary: The toxic effects of polystyrene particles are due to residual styrene monomers, which may lead to ambiguous interpretations of their impact on cells, tissues, and organisms. Therefore, it is important to analyze the residual styrene content. A simple and sensitive UV-vis method was developed to analyze residual styrene and polystyrene concentration. The study found that standard polystyrene particles containing residual monomers exhibited low but significant cytotoxicity, while in-house synthesized polystyrene particles with reduced styrene content did not.

Polystyrene microparticle distribution after ingestion by murine macrophages

Summary: The impact of microplastic particles on organisms is currently under intense research. This study investigated the fate of polystyrene particles ingested by murine macrophages, including their distribution during cell division and possible mechanisms of excretion. The results showed that particle distribution during cell division was cell-specific, and no active excretion of microplastic particles was observed. Additionally, polarized macrophages exhibited higher phagocytic activity and particle uptake compared to non-polarized cells. Furthermore, it was found that submicron particles were co-localized with the endoplasmic reticulum, while 0.5μm particles were occasionally found in endosomes.
