Simultaneous determination of 10 β2-agonists in swine urine using liquid chromatography-tandem mass spectrometry and multi-walled carbon nanotubes as a reversed dispersive solid phase extraction sorbent

A simple and inexpensive pretreatment procedure was developed for 10 (beta(2)-agonists (clenbuterol, ractopamine, salbutamol, bambuterol, penbuterol, tulobuterol, clorprenaline, mabuterol, cimaterol and terbutaline) in swine urine using dispersive solid phase extraction (dSPE) with multi-walled carbon nanotubes (MWCNTs). The sample was analysed after purification by ultra high performance liquid chromatography-positive electrospray ionisation tandem mass spectrometry (UHPLC-ESI-MS/MS). The pH value of the swine urine, extraction time, type and amount of MWCNTs and type of eluent were optimised to increase the sample throughput and sensitivity. The samples were quantified using clenbuterol-D-9, ractopamine-D-6 and salbutamol-D-3 as internal standards. The recoveries of the target compounds from swine urine samples at pH 10.0 were most efficient when using 20 mg of MWCNTs with a 30-50 nm outer diameter and a length of 10-30 mu m, while a mixture of water/methanol (90:10, v/v) with 0.5% formic acid was shown to be the most suitable solvent for desorbing the compounds from the MWCNTs. The proposed method was validated according to the European Commission Decision 2002/657/EC, which determines linearity, specificity, decision limit (CC alpha), detection capability (CC beta), recovery, precision and stability. Studies on the Transmission of a Tigecycline Resistance-Mediating tet(A) Gene Variant from Enterobacter hormaechei via a Two-Step Recombination Process

Summary: This study investigates the transfer and recombination events of a tet(A) variant conferring tigecycline resistance in Enterobacter hormaechei. The results show that the tet(A) variant was present in E. hormaechei G17 and transferred between plasmids in a two-step recombination process. This transfer of resistance genes enhances the dissemination of tigecycline resistance and compromises the effectiveness of treatment for E. hormaechei infection.

A Small Multihost Plasmid Carrying erm(T) Identified in Enterococcus faecalis

Summary: The study aimed to determine the mobile genetic elements involved in the horizontal transfer of erythromycin resistance in Enterococcus faecalis and investigate its transmission ability in heterologous hosts. It was found that a plasmid carrying the erm(T) gene could be transferred to different bacteria and conferred resistance to erythromycin and clindamycin.

Plasmid-Assisted Horizontal Transfer of a Large lsa(E)-Carrying Genomic Island in Enterococcus faecalis

Summary: This study demonstrates the essential role of a pheromone-responsive conjugative plasmid in the horizontal transfer of a lsa(E)-carrying genomic island in Enterococcus faecalis. The presence of this plasmid enables the dissemination of both plasmid-borne resistance genes and chromosomally-located genomic islands. This finding highlights the importance of pheromone-responsive conjugative plasmids in the genomic evolution of E. faecalis, a leading nosocomial pathogen worldwide.

Excision and integration of unconventional circularizable structures involving the erm(B) gene in enterococci

Summary: Traditionally, insertion sequences (ISs) and unconventional circularizable structures (UCSs) have been shown to play a major role in disseminating antimicrobial resistance genes (ARGs) in bacteria. This study identified a novel UCS on plasmid pE508-2 in E. faecalis E508, and demonstrated the excision and integration of UCS involving structures that include the erm(B) gene. The widespread presence of these UCSs in various Gram-positive bacteria highlights its importance in the dissemination of ARGs among bacterial pathogens.

Summary: This study characterized the co-resistance and co-dissemination mechanism of carbapenem and fosfomycin resistance in an Escherichia coli strain isolated from chicken meat. The presence of the blaNDM-5 and fosA3 genes was confirmed, and their transferability to another strain was demonstrated. Whole genome sequencing revealed a novel plasmid carrying the blaNDM-5 and fosA3 genes, and the potential transfer of these genes was identified through translocation units.

Emergence of lnu(C) variant conferring lincomycin resistance in Campylobacter coli of chicken origin

Summary: Lincomycin is frequently used in veterinary medicine and food animal production for treating respiratory and gastrointestinal infections. This study identified a lnu(C) variant in Campylobacter from chickens that confers resistance to lincomycin. The lnu(C) variant showed amino acid substitutions compared to the originally reported lnu(C) from Streptococcus agalactiae. Cloning the lnu(C) variant into lincomycin-susceptible Campylobacter confirmed its function in increasing resistance.

Summary: This study aimed to investigate the transferability of acquired linezolid resistance genes and associated mobile genetic elements in Enterococcus faecalis isolate QZ076. By analyzing the genome and conducting conjugation experiments, it was found that the resistance genes were present on multiple plasmids and a pseudocompound transposon. Furthermore, the transfer of these resistance genes was observed between different bacterial species. This study highlights the potential for the rapid dissemination of acquired linezolid resistance genes and emphasizes the importance of monitoring and controlling their spread.

Summary: This study aimed to determine the molecular mechanisms of linezolid-resistant enterococci (LRE) in swine slaughterhouses in China and analyze the evolutionary dynamics of poxtA-positive E. faecium worldwide. Swine faeces were found to be a significant reservoir for LRE, and optrA and poxtA were the main contributors to linezolid resistance. Phylogenetic analysis revealed the potential global transmission of CC17 poxtA-positive E. faecium between humans and animals.

Summary: This study investigated the presence and location of erm(T) in clinical Streptococcus suis isolates and explored the transmission ability and fitness cost of erm(T)-carrying mobile genetic elements. The results showed that erm(T) was detected on a plasmid and an integrative and conjugative element (ICE) in S. suis isolates, which were transmissible by transformation. The erm(T)-carrying plasmid and ICE posed a fitness cost to the host S. suis isolate.

Breakthrough curve analysis of phosphorylated hazelnut shell waste in column operation for continuous harvesting of lithium from water

Summary: Biosorption using phosphorylated hazelnut shell waste (FHS) is a promising method for lithium removal and recovery. This study investigated the performance of a continuous-flow packed-bed column system using FHS for ultra-low level lithium removal. The results showed that the lihium biosorption capacity of the FHS column was unaffected by the bed height, but decreased with increasing flow rate.

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

An integrated approach for studying the metabolic profiling of herbal medicine in mice using high-resolution mass spectrometry and metabolomics data processing tools

Summary: Analysis of in vivo exposure to traditional Chinese medicine using mass spectrometry is helpful for identifying effective ingredients and developing new drugs. This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles

The method demonstrated good linearity, reproducibility, and recovery, and was successfully applied to analyze 20 wine samples.
