Simultaneous analysis of the main markers of nitrogen status in dairy cow's urine using hydrophilic interaction chromatography and tandem mass spectrometry detection

Microbial protein synthesis and nitrogen balance status in ruminants can be evaluated by the presence of metabolites in urine. This work aims to develop and validate a simple and sensitive method for simultaneous determination of nine markers of nitrogen status in ruminant's urine using hydrophilic interaction liquid chromatography coupled with electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS). The LC-ESI-MS/MS system, operated in multiple reaction monitoring (MRM) in positive mode, was used for determining selected purine (allantoin, uric acid, xanthine, and hypoxanthine) and pyrimidine derivatives (beta-aminobutyric acid and beta-alanine), which are used to estimate rumen microbial protein synthesis - the main source of protein for ruminants. Creatinine, creatine and urea, three other metabolites involved in nitrogen metabolism were also measured by this method. The procedure was based on a simple dilution of urine samples in acetonitrile, followed by LC-ESI-MS/MS analysis. Chromatographic separation was tested with three different columns. A zwitterionic hydrophilic interaction liquid chromatography (ZIC-HILIC) column provided an optimal separation for all metabolites. Precision of the method was typically below 10%, and accuracy was above 90% with the exceptions of allantoin and urea at pH 6 and 3, and beta-alanine at pH 3. Metabolites were stable after 3 months of storage at -20 degrees C, except for xanthine, hypoxanthine and beta-aminobutyric acid that lost up to 48.50 and 39% of initial concentration after only 1 month of storage in acidified urine. This LC-ESI-MS/MS method is more specific, unequivocally detecting target metabolites at lower detection limits than methods using UV detection. The method was suitable for the determination of all metabolites tested. The developed method was subsequently used to compare total and spot urine sampling obtained from dairy cows fed diets with contrasting levels of protein. (C) 2012 Elsevier B.V. All rights reserved.

Metaproteomics Approach and Pathway Modulation in Obesity and Diabetes: A Narrative Review

Summary: Low-grade inflammatory diseases have been found to be associated with metabolic disturbances, such as gut microbiota dysbiosis. Metaproteomics has been used in the past decade to investigate protein composition in specific healthy and pathological conditions. This review applied a rigorous protocol to select 10 metaproteomics studies aiming at investigating obesity and diabetes, providing insights into microbial and human metaproteome alterations and metabolic patterns in individuals affected by diabetes and obesity. However, the lack of metaproteomic data limits objective considerations, and the establishment of specific databases and standardized analysis protocols is needed to further enhance our knowledge on these prevalent diseases.

The host genetics were found to be related to the microbial composition in milk.

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

An integrated approach for studying the metabolic profiling of herbal medicine in mice using high-resolution mass spectrometry and metabolomics data processing tools

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles
