Investigations on the chromatographic behaviour of zwitterionic stationary phases used in hydrophilic interaction chromatography

Two commercial stationary phases possessing a sulfobetaine zwitterionic bonded ligand (ZIC-HILIC and Nucleodur HILIC) were compared under hydrophilic interaction chromatographic (HILIC) conditions. First of all, the separation of 12 model compounds chosen among neurotransmitters and presenting a diversity of ionization states (anionic. cationic and zwitterionic) was studied under varied operating conditions. The effects of the percentage of acetonitrile. ammonium acetate concentration and temperature of the mobile phase were compared on the two columns. Secondly, a generally applicable retention model was established, based on chromatographic retention data (log k) acquired for 76 model compounds. The chosen compounds are small molecules presenting a wide diversity of molecular structures and are relevant to biomedical and pharmaceutical studies. To account for their retention behaviour, a modified version of the solvation parameter model was designed: two additional molecular descriptors were introduced, to account for ionic interactions with anionic and cationic species. The retention equations obtained allow a rationalization of the interactions contributing to retention and separation in the HILIC systems considered. (C) 2011 Elsevier B.V. All rights reserved.

Summary: The development of automatized hyphenated systems is attractive for avoiding sampling error, saving time, and reducing costs. Among these systems, on-line SFE-SFC allows efficient, quick, and eco-friendly extraction, separation, and detection of molecules. The principles of supercritical fluid extraction (SFE) and supercritical fluid chromatography (SFC) are discussed, along with comparisons of partially-supercritical on-line systems before examining the evolution of on-line SFE-SFC over the past decades. Different devices for better understanding coupling challenges and determining advantageous configurations are also examined.

Defining a generic column set for achiral supercritical fluid chromatography applied to pharmaceuticals or natural products

Summary: When starting method development in SFC, the choice of suitable stationary phases can be difficult due to the large number of options available. In this project, 16 columns with diverse stationary phases and polarities were compared to determine their performance. The ranking was based on multiple criteria including the number of eluted compounds, elution time, peak width, peak symmetry, and retention along the gradient time. Columns with high overall scores, such as bare silica gel, propanediol-bonded silica, and pentabromobenzyloxy-bonded silica, were found to be advantageous.

Effect of dilution solvent and injection volume on the analysis of basic hydrophilic therapeutic polypeptide salts with pressurized carbon dioxide mobile phases

Summary: The chromatographic analysis of long-chain hydrophilic therapeutic peptides was explored using pressurized CO2 as the mobile phase. The optimal method was obtained on a Torus 2-PIC column with a gradient elution of 50-90% co-solvent in CO2. UV and mass spectrometry were used to assess the purity and resolution of the major peak and its impurities. Peak distortion was observed in some cases, and thorough examination of dilution solvent and injection volume was conducted to improve peak shape and impurity resolution.

Comparison of the triglyceride composition of vegetable samples with ultra-high efficiency/low-pressure supercritical fluid chromatography - mass spectrometry

Summary: The composition and abundance of triglycerides in vegetable lipid samples were analyzed using ultra-high efficiency / low-pressure supercritical fluid chromatography coupled with mass spectrometry. A total of 94 different triglycerides were identified and quantified, including low-abundance triglycerides and structurally similar isomers. Hierarchical cluster analysis was used to compare the similarity between samples, allowing for comparison of samples from different vegetable sources and the impact of refining treatments on triglyceride content.

Summary: Plant seeds, especially milk thistle seeds, contain valuable nonpolar and polar compounds with potential biological activity. This study developed an online method called supercritical fluid extraction (SFE)-supercritical fluid chromatography (SFC) to extract and analyze both types of compounds. Different transfer modes were compared, and the challenges of analyzing nonpolar and polar compounds were discussed. The versatility of online SFE-SFC and its potential in analyzing both types of compounds in a single sample were explored.

Breakthrough curve analysis of phosphorylated hazelnut shell waste in column operation for continuous harvesting of lithium from water

Summary: Biosorption using phosphorylated hazelnut shell waste (FHS) is a promising method for lithium removal and recovery. This study investigated the performance of a continuous-flow packed-bed column system using FHS for ultra-low level lithium removal. The results showed that the lihium biosorption capacity of the FHS column was unaffected by the bed height, but decreased with increasing flow rate.

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

An integrated approach for studying the metabolic profiling of herbal medicine in mice using high-resolution mass spectrometry and metabolomics data processing tools

Summary: Analysis of in vivo exposure to traditional Chinese medicine using mass spectrometry is helpful for identifying effective ingredients and developing new drugs. This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles

Summary: A quantitative analytical method was developed and validated in this study to determine the concentrations of 81 aroma-relevant wine volatiles. The method utilizes stir bar sorptive extraction and multidimensional gas chromatography with flame ionization detector or mass spectrometer. The method demonstrated good linearity, reproducibility, and recovery, and was successfully applied to analyze 20 wine samples.
