A number of N,N-dialkylated tryptamines show psychoactive properties in man which resulted in a renewed interest in psychopharmacological research. Attempts to manufacture these derivatives are increasing within a clandestine environment, where literature procedures are adapted and information is exchanged on the Internet. One such example is based on the thermolytic decarboxylation of tryptophan to tryptamine as the precursor to psychoactive derivatives. This procedure was proposed to make use of household solvents such as turpentine substitute and white spirit to facilitate decarboxylation. Discussions on websites also suggested the catalytic use of natural oils in order to accelerate these reactions. In this research, the analytical characterization of this preparation procedure was carried out using gas chromatography-ion trap single and tandem stage mass spectrometry in electron and chemical ionization mode that led to the identification of previously unreported 1-mono and 1,1-disubstituted tetrahydro-beta-carboline (THBCs) by-products. The tryptamine product and several THBC by-products were determined quantitatively and a fingerprint analysis of the crude products allowed for the differentiation between the essential oil catalysts involved as indicated by the presence of tetrahydro-beta-carbolines and their imine intermediates. Summary: This study characterized a novel lysergamide N,N-diethyl-1-propanoyl-6-(prop-2-en-1-yl)-9,10-didehydroergoline-8 beta-carboxamide (1P-AL-LAD) using various analytical methods. In vitro metabolism studies confirmed that 1P-AL-LAD acted as a prodrug and converted to AL-LAD as the most abundant metabolite. 1P-AL-LAD induced dose-dependent head twitch response (HTR) in mice, similar to other hallucinogenic substances.

Formation, Characterization, and Occurrence of β-Carboline Alkaloids Derived from α-Dicarbonyl Compounds and L-Tryptophan

Summary: This study discovers that new beta-carbolines can be formed when L-tryptophan reacts with alpha-dicarbonyl compounds. The formation of these beta-carbolines increases under acidic conditions and with increasing temperature. These alpha-dicarbonyl-derived beta-carbolines are present in foods, formed during heating processes, and can be considered as advanced glycation end products.

Tadalafil Rescues the p.M325T Mutant of Best1 Chloride Channel

Identification, Formation, and Occurrence of Perlolyrine: A beta-Carboline Alkaloid with a Furan Moiety in Foods

Summary: This research identifies and characterizes perlolyrine, a beta-carboline compound with a furan moiety, and reveals its formation mechanism and occurrence in various foods. Perlolyrine is formed from the reaction of L-tryptophan with 3-deoxyglucosone, an intermediate of carbohydrate degradation, under acidic conditions and temperatures in the range of 70-110 degrees C. It is widely present in processed foods and is ingested daily through the diet.

Computational Investigation of Mechanisms for pH Modulation of Human Chloride Channels

Summary: Many transmembrane proteins are modulated by intracellular or extracellular pH. Mutagenesis guided by amino-acid conservation and structure can be used to investigate pH dependence, but prediction of pKas can streamline this process. The hypothesis of modulation of calcium/proton coupling and pH-dependent activity through the extent of structural relaxation applies to various proteins, including bestrophin 1, TMEM16A, calmodulin, and calbindin D9k. Predicting altered pH dependence for mutations in ion channel disorders can support experimental studies and clinical intervention.

Physiological and endocrine measures were also included. In serum and urine, MDAI could be detected for at least 4 and 6 days.

Breakthrough curve analysis of phosphorylated hazelnut shell waste in column operation for continuous harvesting of lithium from water

Summary: Biosorption using phosphorylated hazelnut shell waste (FHS) is a promising method for lithium removal and recovery. This study investigated the performance of a continuous-flow packed-bed column system using FHS for ultra-low level lithium removal. The results showed that the lihium biosorption capacity of the FHS column was unaffected by the bed height, but decreased with increasing flow rate.

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

An integrated approach for studying the metabolic profiling of herbal medicine in mice using high-resolution mass spectrometry and metabolomics data processing tools

Summary: Analysis of in vivo exposure to traditional Chinese medicine using mass spectrometry is helpful for identifying effective ingredients and developing new drugs. This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles

The method demonstrated good linearity, reproducibility, and recovery, and was successfully applied to analyze 20 wine samples.
