Weak cation-exchange restricted-access material for on-line purification of basic drugs in plasma

A methylcellulose-immobilized weak cation-exchange (MC-WCX) silica-based restricted-access material (RAM) was developed. The MC-WCX consists of an MC outer surface and 2-carboxyethyl phase internal surface, allowing for direct analysis of basic drugs in plasma. The retention properties of the MC-WCX were evaluated for sulpiride, quinidine, ranitidine, and desipramine. The MC-WCX retained model drugs by cation-exchange, and retained drugs were eluted with the mobile phase containing small amount of acids or salts compared with the MC strong cation-exchanger (MC-SCX). These results indicated the ease of use of the MC-WCX solid-phase extraction (SPE) column when coupled to a reversed-phase analytical column in column-switching high-performance liquid chromatography (HPLC), and various detection principals. Further direct analysis of model drugs in plasma using the MC-WCX SPE column in a column-switching HPLC system successfully performed with sufficient recovery. Summary: A non-destructive discrimination method using Raman spectroscopy and X-ray CT was developed to differentiate between crystal polymorphs of ranitidine in tablet formulations. X-ray CT imaging revealed varying densities, sizes, and shapes of crystal particles within the same batch, suggesting its potential as a unique method for quantitative physicochemical characterization of crystal polymorphs and solid dosage forms.

Chemical imaging analysis of active pharmaceutical ingredient in dissolving microneedle arrays by Raman spectroscopy

Summary: This study developed a quality evaluation method for dissolving microneedle arrays (DMNAs) and explored the spatial distribution of drugs in DMNAs using Raman spectroscopy mapping. The analysis revealed the crystallization forms of diclofenac sodium (DCF) in the polymer films and the linear relationship between drug content and characteristic peak intensity in the films. Raman imaging demonstrated a homogeneous dispersion of small DCF crystals in the films, as well as the distribution of DCF on the surface of the DMNA needle.

A method for purifying nanoparticles using cationic modified monoliths and aqueous elution

Summary: In this study, three types of cationic modified silica monoliths were used to separate and purify nanoparticles, and the nanoparticles were separated from other substances by changing the pH and concentration of the eluent. The results showed that this method can extract nanoparticles in a short time with only low-speed centrifugation and has a high recovery efficiency.

Evaluation of Drug Sorption on Laboratory Materials with Abraham Solvation Parameters of Drugs and its Prevention

Summary: Drug sorption on laboratory materials can affect the performance of analytical methods and lead to unreliable data. By using a linear free energy relationship (LFER) model with Abraham solvation parameters, drug sorption can be characterized and evaluated to prevent it. The physicochemical properties of drugs, rather than the materials used, play a key role in sorption, and additives like organic solvents can alter the rate of drug sorption. Equations have been developed to assess drug sorption extent on glass and polypropylene, and the LFER model is valuable for studying sorption properties in drug discovery research and other biochemical fields.

Investigation of Foreign Particles in Moderna COVID-19 Vaccine

Folded, undulating, and fibrous doxorubicin sulfate crystals in liposomes

Summary: High-resolution cryogenic transmission electron microscopy (cryo-TEM) revealed that doxorubicin sulfate crystals in liposomes (prepared by remote loading with ammonium sulfate) formed folded, undulating, and fibrous structures with a diameter of approximately 2.4 nm. An undulating, fibrous crystal considered to be undergrowth, as well as bundles of fibrous crystals, were observed in doxorubicin-loaded liposomes. This explains the formation of various shaped doxorubicin sulfate crystals, such as curved, U-shaped, or circular, in addition to the cylinder and/or rod-like crystals reported in the literature. Liposomes without crystals contained high-density aqueous phases, indicating that doxorubicin is loaded remotely and remains as a solute without precipitation.

Although advancements in liquid chromatography have improved efficiency, difficulties in separating similar impurities with high-performance liquid chromatography (HPLC) still remain. Bayesian inference is applied to develop an algorithm that assigns confidence intervals to quantitative data of each analogous substance using the MCR-ALS method.

Algorithm for the early prediction of drug stability using bayesian inference and multiple measurements: Application for predicting the stability of silodosin tablets

Summary: The stability of APIs and formulations is crucial in the pharmaceutical industry, affecting research and development feasibility, development period, and costs. To predict early-stage formulation stability accurately, we developed an algorithm combining Bayesian inference and the conventional Arrhenius equation, providing a narrow confidence interval even with data variations. Our method enables the reasonable prediction of long-term drug stability and contributes to pharmaceutical product development worldwide.

Breakthrough curve analysis of phosphorylated hazelnut shell waste in column operation for continuous harvesting of lithium from water

Summary: Biosorption using phosphorylated hazelnut shell waste (FHS) is a promising method for lithium removal and recovery. Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

An integrated approach for studying the metabolic profiling of herbal medicine in mice using high-resolution mass spectrometry and metabolomics data processing tools

Summary: Analysis of in vivo exposure to traditional Chinese medicine using mass spectrometry is helpful for identifying effective ingredients and developing new drugs. This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles

Summary: A quantitative analytical method was developed and validated in this study to determine the concentrations of 81 aroma-relevant wine volatiles. The method demonstrated good linearity, reproducibility, and recovery, and was successfully applied to analyze 20 wine samples.
