SUMO2/3 conjugation is an endogenous neuroprotective mechanism

Small ubiquitin-like modifier (SUMO) 2/3 but not SUMO1 conjugation is activated after transient cerebral ischemia. To investigate its function, we blocked neuronal SUMO2/3 translation through lentiviral microRNA delivery in primary cortical neurons. Viability was unaffected by SUMO2/3 silencing unless neurons were stressed by transient oxygen-glucose deprivation (OGD). Both 15 and 45 minutes of OGD were tolerated by control microRNA-expressing neurons but damaged >60% of neurons expressing SUMO2/3 microRNA. Damaging OGD (75 minutes) increased neuronal loss to 54% (control microRNA) and to 99% (SUMO2/3 microRNA). This suggests that activation of SUMO2/3 conjugation is an endogenous neuroprotective stress response. Summary: This study evaluated the ability of quantitative DWI and FLAIR to predict DWI-FLAIR mismatch in ischemic stroke. The results showed that the agreement between quantitative parameters and DWI-FLAIR mismatch was insufficient and there was a moderate correlation between the two quantitative parameters. The interquartile range rDWI SI was found to be associated with excellent functional outcome.

Summary: There is evidence of an association between levels of high-sensitivity cardiac troponin T (hs-cTnT) and cognitive decline in older men, as measured by the Digit Symbol Substitution Test (DSST). This association remained significant even after adjusting for age, sex, education, and cardiovascular risk factors. However, there was no significant association between hs-cTnT and different cognitive domains at baseline.

Summary: This study found that elevated cardiac biomarkers, such as high-sensitivity cardiac troponin T (hs-cTnT) and brain natriuretic peptide (BNP), in acute stroke patients are strongly associated with pathological findings on cardiac magnetic resonance imaging (CMR). The elevation of these biomarkers may help identify patients who require a more comprehensive cardiology work-up.

Zfp580 inactivation as a new therapeutic target to enhance recovery after stroke in mice

Summary: Paracrine cerebral Interleukin 6 (Il6) plays a role in stroke recovery, but systemic Il6 elevation may worsen outcome. Lithium modulates Il6 responses and improves stroke outcome, but has adverse effects. Zincfinger protein 580 (Zfp580) mediates the effects of lithium on Il6 signaling without adverse effects, improving cognitive and motor function. Inactivation of Zfp580 has positive effects on multiple key mechanisms, making it a potentially more specific and effective treatment target for stroke recovery than lithium. Zfp580 inhibitors must be developed to fully assess its potential.

Deep learning-based automated lesion segmentation on mouse stroke magnetic resonance images

Summary: This study developed a cost-efficient and scalable human cell line for the production of immunogenic SARS-CoV-2 virus-like particles (VLPs). The modular design of the VLP production platform allows for rapid adaptation to new variants. The generated VLPs contained all structural proteins, exhibited specific reactivity with serum from naturally infected individuals, and demonstrated stability at low temperatures. Mice immunized with these VLPs produced neutralizing antibodies against the SARS-CoV-2 spike protein. The findings highlight the efficient, scalable, and adaptable nature of this production method, which holds therapeutic potential.
