GKN2 Contributes to the Homeostasis of Gastric Mucosa by Inhibiting GKN1 Activity

Gastrokine 1 (GKN1) plays an important role in maintaining gastric mucosa integrity. Here, we investigated whether gastrokine 2 (GKN2) contributes to the homeostasis of gastric epithelial cells by regulating GKN1 activity. We analyzed cell viability, proliferation, and death in AGS cells transfected with GKN1, GKN2, GKN1 plus GKN2 using MTT, BrdU incorporation, and apoptosis assays, respectively. In addition, the expression levels of the cell cycle- and apoptosis-related proteins, miR-185, DNMT1, and EZH2 were determined. We also compared the expression of GKN1, GKN2, and CagA in 50 non-neoplastic gastric mucosae and measured GKN2 expression in 169 gastric cancers by immunohistochemistry. GKN2 inhibited anti-proliferative and pro-apoptotic activities, miR-185 induction, and anti-epigenetic modifications of GKN1. There was a positive correlation between GKN1 and GKN2 expression (P=0.0074), and the expression of GKN1, but not GKN2, was significantly lower in Helicobacter pylori CagA-positive gastric mucosa (P=0.0013). Interestingly, ectopic GKN1 expression in AGS cells increased GKN2 mRNA and protein expression in a time-dependent manner (P=0.01). Loss of GKN2 expression was detected in 126 (74.6%) of 169 gastric cancers by immunohistochemical staining and was closely associated with GKN1 expression and differentiation of gastric cancer cells (P=0.0002 and P=0.0114, respectively). Overall, our data demonstrate that in the presence of GKN2, GKN1 loses its ability to decrease cell proliferation, induce apoptosis, and inhibit epigenetic alterations in gastric cancer cells. Re-identification and Characterization of Multidrug-Resistant Corynebacterium haemomassiliense-Like Organisms from Humans

Summary: Recently, there has been an increase in the number of reported infections caused by non-diphtheriae Corynebacterium spp. in patients, as well as the discovery of novel species of Corynebacterium isolated from humans. We report two cases of human infections caused by Corynebacterium haemomassiliense-like organisms, which were initially unidentified by MALDI-TOF MS analysis. Further analysis using three housekeeping genes and phenotypic features revealed their close relation to the recently described species C. haemomassiliense. These strains were found to be multidrug-resistant but susceptible to vancomycin, meropenem, and linezolid. Our report suggests that these recently described Corynebacterium species may not be limited to a specific region, highlighting the difficulty in classifying the genus Corynebacterium.

Summary: Due to the broad spread, incidence, and genetic divergence of enteroviruses (EVs), it has been challenging to deal with this virus that causes severe human diseases. Therefore, an efficient and universal cloning system for the reverse genetics of highly divergent EVs contributes to an understanding of the viral pathology and molecular mechanisms of evolution.

Association of scrub typhus with incidence of dementia: a nationwide population-based cohort study in Korea

Summary: This study used large population-based cohort data from South Korea to investigate the association between scrub typhus and dementia. The results showed that a history of scrub typhus infection in old age is significantly associated with an increased risk of dementia, especially Alzheimer's disease. Therefore, prevention and appropriate treatment of scrub typhus should be emphasized as a measure for dementia prevention.

Risk of breakthrough SARS-CoV-2 infection and clinical outcomes among vaccinated patients with type 2 diabetes

Summary: This study aimed to explore the risk of breakthrough infection among patients with type 2 diabetes and the risk of severe clinical outcomes after SARS-CoV-2 infection according to vaccination status. The results showed that patients with type 2 diabetes were more likely to experience breakthrough infections, but fully vaccinated patients had a lower risk of adverse clinical outcomes after SARS-CoV-2 infection.

The population is ageing at an unprecedented pace in the Asia-Pacific (APAC) region, with the number of persons older than 65 years set to double by 2050 to around 1.3 billion. This review provides an overview of the challenges to adult vaccination in APAC, drivers to increase vaccination coverage, vaccination insights gained through the COVID-19 pandemic, and potential measures to increase the uptake of adult vaccines in the region.

A Rapid Method for Generating Infectious SARS-CoV-2 and Variants Using Mutagenesis and Circular Polymerase Extension Cloning

This method is valuable for studying the molecular characteristics of emerging variants and for the development and testing of vaccines and antivirals.

Ri sk of mortality and clinical outcomes associated with healthcare delay among patients with tuberculosis

Summary: This study aimed to measure healthcare delay and its association with clinical outcomes among tuberculosis (TB) patients from 2013 to 2018. The results showed that the mean healthcare delay was 42.3 days, and it was associated with increased risk of all-cause mortality, pneumonia, and mechanical ventilation use. The findings suggest the need for attention from authorities and healthcare professionals to timely treatment in order to reduce the preventable burden caused by TB.

Summary: This article presents a 140GHz 4-element RF beamforming phased-array receiver (RX) implemented in 22nm FDSOI CMOS. The receiver utilizes a single-side-band architecture and achieves image rejection ratios of >25dB and >20dB across 4GHz and 8GHz channel bandwidth centered at 7GHz intermediate frequency (IF). The RX consumes 480mW DC power, has a noise figure of <10dB from 135 to 147 GHz, and achieves data rates of up to 32Gb/s and 24Gb/s in probe and over-the-air tests.

HZ in IC/AID patients leads to higher medical costs.
