Depression and type 2 diabetes: Cortisol pathway implication and investigational needs

Depression and type 2 diabetes (T2D) are clinically associated and the causes of the association are still under investigation. We aimed at identifying what is known about the stress response and cortisol pathway and the clinical association of depression and T2D, and at hypothesizing the link of the association. In this review, we report independent studies on stress response, cortisol pathway, depression, T2D, and independent studies on stress and cortisol pathway in depression, and in T2D. We focus and integrate the stress and cortisol pathway hypothesis to explain the clinical association of depression and T2D. We hypothesize that the corticotropin-releasing hormone receptors are one of the missing linking factor of the cortisol pathway underlying the clinical association of depression and T2D. This study found that MC2R and MC5R genes are associated with MDD, while MC1R, MC2R, MC3R, and MC4R genes are associated with T2D. These findings provide important insights into the role of melanocortin receptor genes in the comorbidity of MDD and T2D.

Comorbidity of Novel CRHR2 Gene Variants in Type 2 Diabetes and Depression

Summary: Variants of the CRHR2 gene are associated with the risk of depression and type 2 diabetes. This study identified novel risk variants related to depression and type 2 diabetes in 212 Italian families. The findings suggest that CRHR2 plays a stronger role in the risk of depression than in the risk of type 2 diabetes.

Many genetic risk factors are not yet known, but the mineralocorticoid receptor gene (NR3C2) has been identified as a potential risk gene for PCOS. This study found several novel risk variants in NR3C2 that are associated with the development of PCOS.

Summary: Polycystic ovarian syndrome (PCOS) is a complex heterogeneous disorder characterized by hyperandrogenism, irregular menses, and subfertility and often accompanied by other related comorbid disorders such as insulin resistance, obesity, and type 2 diabetes. We found 18 novel risk variants of the NR3C2 gene significantly linked to and/or associated with the risk of PCOS. However, our findings need to be replicated in other ethnic groups in order to reach more solid conclusions.

PSMD9??????? is linked to T2D age of onset, years of isolated and combined insulin therapy, irregular menses, and hot flashes

Summary: This study analyzed the link between PSMD9 rs74421874 (IVS3+nt460-G > A), rs3825172 (IVS3+nt437-C > T), and rs14259 SNPs and the quantitative traits of type 2 diabetes (T2D) and the qualitative phenotypes of irregular menses, couple infertility, and menopausal hot flashes. The results showed that PSMD9 is linked to irregular menses, menopausal hot flashes, T2D age of onset, years of combined oral and insulin therapy, and only insulin therapy.

Linkage and association of novel DRD2 variants to the comorbidity of type 2 diabetes and depression
