Sequential IL-23 and IL-17 and increased Mmp8 and Mmp14 expression characterize the progression of an experimental model of periodontal disease in type 1 diabetes

Molecular mechanisms responsible for periodontal disease (PD) and its worsening in type 1 Diabetes Mellitus (DM1) remain unknown. Cytokine profile and expression levels of collagenases, Mmp14, and tissue inhibitors were determined, as were the numbers of neutrophils and macrophages in combined streptozotocin-induced DM1 and ligature-induced PD models. Increased IL-23 (80-fold) and Mmp8 expression (25-fold) was found in DM1. Ligature resulted in an IL-1 beta/IL-6 profile, increased expression of Mmp8, Mmp13, and Mmp14 (but not Mmp1), and transient expression of Timp1 and Reck in non-diabetics. PD in DM1 involved IL-1 beta (but not IL-6) and IL-23/IL-17, reduced IL-6 and IL-10, sustained Mmp8 and Mmp14, increased Mmp13 and reduced Reck expression in association with 20-fold higher counts of neutrophils and macrophages. IL-23 and Mmp8 expression are hallmarks of DM1. In association with the IL-1/IL-6 (Th1) response in PD, one found a secondary IL-17 (Th17) pathway in non-diabetic rats. Low IL-6/TNF-a suggest that the Th1 response was compromised in DM1, while IL-17 indicates a prevalence of the Th17 pathway, resulting in high neutrophil recruitment. Mmp8, Mmp13, and Mmp14 expression seems important in the tissue destruction during PD in DM1. PD-associated IL-1/IL-6 (Th1), IL-10, and Reck expression are associated with the acute-to-chronic inflammation transition, which is lost in DM1. Validation in a Brazilian population of gene markers of periodontitis previously investigated by GWAS and bioinformatic studies

Summary: Genetic markers of susceptibility to periodontitis were validated in a Brazilian population, with SNPs in the NPY, IL37, and NCR2 genes associated with moderate or severe periodontitis, and the TLR9 marker associated with lower risk of severe periodontitis in men. Sex- and smoking-specific effects were observed for these SNPs, highlighting the genetic predisposition to periodontitis.

Novel inhibitor of hematopoietic cell kinase as a potential therapeutic agent for acute myeloid leukemia

Summary: MDS and AML are hematological disorders with high relapse risk and poor survival. This study investigated the effects of a novel HCK inhibitor, iHCK-37, in combination with 5-Azacytidine or Cytarabine on leukemia cells. The combination treatment showed additive effects and reduced oncogenic pathway activation. In addition, iHCK-37 selectively reduced the number of MDS and AML cells without affecting normal cells. These findings suggest that iHCK-37 may have potential as a targeted therapy for leukemia.

Reduced blood pressure in sickle cell disease is associated with decreased angiotensin converting enzyme (ACE) activity and is not modulated by ACE inhibition

Summary: This study investigated the association between blood pressure and renin-angiotensin system components in sickle cell disease (SCD) patients and mice, as well as the effects of angiotensin converting enzyme (ACE) inhibitor on blood pressure and renin-angiotensin system in SCD mice. The results showed that SCD patients and mice had lower diastolic and systolic blood pressure compared to controls, and the reduced blood pressure was associated with increased renin and markers of kidney damage in SCD. ACE concentrations and ACE enzyme activity were significantly decreased in SCD. The administration of enalapril, an ACE inhibitor, lowered blood pressure, plasma angiotensin II, and organ ACE activity in control mice, but had no additional effect on SCD mice. However, plasma angiotensin II and renin levels were significantly higher in enalapril-treated SCD mice than in treated control mice.

Galanin mediates tumor-induced immunosuppression in head and neck squamous cell carcinoma

Summary: The study found that galanin secreted by head and neck squamous cell carcinoma (HNSCC) cells exhibits immune-suppressive and pro-tumoral effects. Galanin decreases cell proliferation and expression of pro-inflammatory cytokines, while increasing apoptosis and expression of pro-tumoral cytokines/growth factors. It also suppresses T cell activation and anti-tumoral phenotypes. High expression of galanin is associated with reduced overall survival in HNSCC patients.

Influence of tumor cell-derived TGF-β on macrophage phenotype and macrophage-mediated tumor cell invasion

Summary: In oral squamous cell carcinoma, macrophages in the tumor microenvironment play a significant role in prognosis and disease survival. This study focuses on the effects of tumor cell-secreted TGF-beta on macrophage phenotype and macrophage-facilitated tumor invasion. The results show that blocking the effects of TGF-beta can affect macrophage phenotype and impact tumor cell invasion and chemotaxis differently.

Stress induced phosphoprotein 1 overexpression controls proliferation, migration and invasion and is associated with poor survival in oral squamous cell carcinoma

Summary: In this study, the expression and impact of stress induced phosphoprotein 1 (STIP1) in oral squamous cell carcinoma (OSCC) were investigated. It was found that STIP1 was highly expressed in OSCC, and its overexpression was associated with shorter survival and higher risk of recurrence. Knockdown of STIP1 reduced proliferation, migration, and invasion of tumor cells, and decreased the number of metastases in a zebrafish model. Furthermore, STIP1 was inversely expressed with miR-218-5p, and transfection of miR-218-5p mimics decreased STIP1 levels and inhibited tumor cell proliferation, migration, and invasion.

Progress in kidney transplantation: The role for systems immunology

Summary: The development of systems biology has led to significant advancements in translational research and personalized medicine. By combining multidisciplinary approaches and novel techniques, it is now possible to extract and analyze vast amounts of data from human samples, even when they are limited in volume and source. These advancements have made the application of a multi-omics approach more accessible outside of specialized centers. In this review, the focus is on biological techniques that are improving our understanding of the immune system and its interactions, with specific emphasis on their potential applications in transplant immunology. The review covers advancements in techniques, initial applications, and future developments in the field.

Comparison of monocytic cell lines U937 and THP-1 as macrophage models for in vitro studies

Summary: Understanding macrophage biology is important for understanding various biological processes and developing new immunomodulatory strategies. This study compared the response of macrophages derived from THP-1 and U937 cells to different polarizing conditions, and found that they have different tendencies towards M1 and M2 phenotypes, with THP-1-derived macrophages being more responsive to M1 stimuli.

Cytotoxicity and bioactive potential of new root repair materials for use with BMP-2 transfected human osteoblast cells

Summary: Modified formulations of calcium silicate repair materials with additives have been developed to enhance handling, consistency, biocompatibility, and bioactivity. In this study, the effects of these modified materials on human osteoblastic cells were evaluated. The results showed that the modified cements did not have cytotoxic effects and promoted cell viability, gene expression, and mineralization processes.

Spatial and temporal VEGF receptor intracellular trafficking in microvascular and macrovascular endothelial cells
