Summary: This study demonstrates that alpha-methylene-gamma-lactams can serve as tunable surrogates for highly reactive alpha-methylene-gamma-lactones, with the reactivity of the alpha-methylene modulated by the N substituent. Density functional theory calculations using the M06-2X functional with the SMD solvation model and methyl thiolate as a model nucleophile reliably predict the relative reactivities of the alpha-methylene-gamma-lactams. Quasiharmonic approximations further improve the agreement between experimental results and computational predictions.

Summary: This study investigates the effect of N-glycosylation on the biochemical function of DSPA alpha 2. Mutating the N-glycosylation sites in DSPA alpha 2 resulted in reduced plasminogen activation and fibrin sensitivity. N-glycosylation did not affect the pH resistance or heat resistance of DSPA alpha 2.

This study presents a novel workflow that combines deglycosylation and proteolytic digestion using suspension trapping, without adding extra time to the sample preparation process. The proposed approach improves identification of potential N-glycosylated peptides and can aid in the discovery of known and potential glycoproteins.

Summary: The comprehensive characterization of N-glycans is crucial for the discovery of potential biomarkers and the diagnosis and therapy of diseases. In this study, a porous graphitized carbon biomaterial was designed and fabricated for the effective capture of N-glycans. The biomaterial exhibited excellent performance in the capture of standard N-glycans and was successfully applied in profiling the difference of N-glycans during diabetes progression, discovering potential biomarkers for accurate diagnosis.

Summary: In this study, the advantage of phenylhydrazine labeling for the detection and discrimination of sialic acid linkages was demonstrated. The results showed that phenylhydrazine-labeled sialylated oligosaccharides can be easily recognized and produce favorable fragmentation patterns during mass spectrometry experiments. A comparison of two esterification methods showed that methylation exhibited slightly higher linkage specificity. The simplicity and effectiveness of this methodology were confirmed using model compounds and serum samples.

Summary: This article describes two key components of the N-glycosylation pathway in the halophilic archaea Halobacterium salinarum. The roles of VNG1053G and VNG1054G, two proteins encoded by genes clustered together with a set of genes demonstrated to encode N-glycosylation pathway components, were investigated. It was determined that VNG1053G is the glycosyltransferase responsible for adding the linking glucose, while VNG1054G is the flippase that translocates the lipid-bound tetrasaccharide across the plasma membrane. The absence of VNG1053G or VNG1054G compromises cell growth and motility in Hbt. salinarum, highlighting their importance in N-glycosylation.
