Ablation of bone cells by electroporation

Short intense electrical pulses transiently increase the permeability of the cell membrane, an effect known as electroporation. This can be combined with antiblastic drugs for ablation of tumours of the skin and subcutaneous tissue. The aim of this study was to test the efficacy of electroporation when applied to bone and to understand whether the presence of mineralised trabeculae would affect the capability of the electric field to porate the membrane of bone cells. Different levels of electrical field were applied to the femoral bone of rabbits. The field distribution and modelling were simulated by computer. Specimens of bone from treated and control rabbits were obtained for histology, histomorphometry and biomechanical testing. After seven days, the area of ablation had increased in line with the number of pulses and/or with the amplitude of the electrical field applied. The osteogenic activity in the ablated area had recovered by 30 days. Biomechanical testing showed structural integrity of the bone at both times. Electroporation using the appropriate combination of voltage and pulses induced ablation of bone cells without affecting the recovery of osteogenic activity. It can be an effective treatment in bone and when used in combination with drugs, an option for the treatment of metastases.

Antiosteoporotic Nanohydroxyapatite Zoledronate Scaffold Seeded with Bone Marrow Mesenchymal Stromal Cells for Bone Regeneration: A 3D In Vitro Model

Summary: In this study, zoledronate functionalized hydroxyapatite nanocrystals (HAZOL) were found to promote the adhesion, viability, metabolic activity, and osteogenic commitment of human bone marrow derived mesenchymal stromal cells (hMSCs). The scaffolds supported cell growth and enhanced osteogenic differentiation and gene expression of hMSCs. These results suggest that zoledronate functionalized scaffolds may have a positive influence on bone fracture healing.

Primary High-Grade Myxoid Liposarcoma of the Extremities: Prognostic Factors and Metastatic Pattern

Summary: This study analyzed the history and treatment outcomes of localized, high-grade myxoid liposarcoma (MLS) of the extremities. The results showed that patients with tumors in the extremities had a good overall survival rate and local recurrence-free survival rate. Tumor sites and surgical margins were not significant risk factors for survival, while tumor size was a significant predictor of metastasis-free survival and disease-free survival.

Summary: Pulsed electromagnetic fields (PEMFs) are a type of biophysical stimulation that have shown promising results in improving bone regeneration and preventing bone loss. However, there is limited understanding of the underlying biological mechanisms and the effects on bone healing. The variability in the characteristics of electromagnetic fields and the responsiveness of cells involved in bone healing pose challenges in drawing definitive conclusions. This review summarizes the current research on PEMF stimulation in animal models with bone impairment, highlighting the need for standardized experimental guidelines and further controlled trials.

Benefits of Applying Nanotechnologies to Hydrogels in Efficacy Tests in Osteoarthritis Models-A Systematic Review of Preclinical Studies

Sensitivity, Specificity, and Predictive Values of Tru-Cut® Biopsy in Grading Primary Localized Myxoid Liposarcomas of the Extremities

Summary: Biopsy plays a crucial role in diagnosing myxoid liposarcoma (MLs) due to the significance of histological grade in determining the appropriate treatment. This retrospective study aimed to assess the accuracy of Tru-cut(R) biopsy (TCB) in diagnosing MLs and its impact on patient survival in case of misdiagnosis. The study found that TCB diagnosis may differ from surgical specimens, with a histological grade concordance rate of 64% (Kappa 0.30). Neoadjuvant treatments were associated with pathological downgrading, but misdiagnosis did not affect prognosis. In patients not treated with neoadjuvant therapy, TCB showed a sensitivity of 57% and specificity of 100%.

Unexpected Absence of Skeletal Responses to Dietary Magnesium Depletion: Basis for Future Perspectives?

Summary: This study investigated the effects of a Mg-deficient diet on the bone of 8-weeks-old C57BL/6J male mice using a histological approach. The results showed that mice receiving the Mg-deficient diet had reduced body weight gain and serum Mg concentration, while the expression of kidney magnesiotropic genes tended to increase, indicating an adaptive response to Mg depletion. However, there were no significant differences in bone density and growth plate thickness, which partially differed from previous studies and might be attributed to experimental protocol differences or genetic factors.

Gender-Specific Differences in Human Vertebral Bone Marrow Clot

Summary: Recently, our group studied the use of vertebral bone marrow (vBMA) clot as a cell therapy for spinal fusion. We compared the properties of vBMA clots and derived vertebral mesenchymal stem cells (MSCs) from female and male patients who underwent spinal fusion procedures and received vBMA clot treatment. The results showed that vBMA clots and MSCs from males had higher expression of growth factors (GFs) and greater osteogenic and chondrogenic potential compared to females. The findings also revealed differences in the gene expression profiles of MSCs derived from vBMA clots between female and male donors.

A Planned Multidisciplinary Surgical Approach to Treat Primary Pelvic Malignancies

Summary: The pelvic anatomy presents challenges to orthopedic surgeons due to the large size and close proximity to vital structures of pelvic sarcomas. This study reports a systematic multidisciplinary surgical approach to treating pelvic sarcomas, in which 17 patients were included and treated using a combination of expertise from orthopedic oncology and other surgical specialties. Hindquarter amputation was performed on seven patients, while excision of the tumor was done for 10 patients. Bone defects were reconstructed in six patients using a custom-made 3D-printed pelvic prosthesis. Thirteen patients experienced complications. Well-organized multidisciplinary collaborations are crucial for managing pelvic sarcoma patients and should be conducted in specialized centers. A multidisciplinary surgical approach is essential for achieving successful outcomes and adequate margins.

Monetite vs. Brushite: Different Influences on Bone Cell Response Modulated by Strontium Functionalization

Summary: This study investigated the influence of strontium substitution on the structures of monetite and brushite, and its effect on bone cell response. The results showed that strontium substitution caused changes in cell parameters and morphology, and the release of ions was greater in brushite compared to monetite. The study also found that brushite materials had a more pronounced effect on osteoblast differentiation and osteoclast inhibition.
