Characterization of a Trichoplusia ni hexamerin-derived promoter in the AcMNPV baculovirus vector

The promoter sequences of the encoding genes for the three most abundant hexamerins of the Lepidoptera Trichoplusia ni were isolated and cloned into the Autographa californica multiple nucleopolyhedrovirus (AcMNPV)-derived baculovirus expression vector. From the sequences analyzed, the DNA region driving the expression of the Basic juvenile hormone-suppressible protein 2 (BJHSP-2), denominated pB2, presented the highest promoter strength in the context of the baculovirus vector in Sf21 insect cells. This promoter activity occurred earlier in baculovirus-infected cells than that achieved by a conventional polyhedrin promoter (polh), but surprisingly stopped at 48 h post-infection. A mapping of pB2 essential promoter elements determined that a region of about 400 bp, denominated pB2(9), retained and even increased the transcriptional activity with respect to the parental full-length sequence. Finally, several chimeric combinations of the insect-derived pB2 with the virus-derived conventional polh or p10 promoters were constructed and incorporated into an AcMNPV baculovirus vector. The pB2-p10 combination showed increased recombinant protein expression at early times post-infection and similar expression levels at very late times post-infection in Sf21 cells with respect to conventional late promoters. To the best of our knowledge, pB2 is the first promoter isolated from the Lepidoptera T. ni, the natural host of AcMNPV, to be assayed in a baculovirus expression vector. (C) 2013 Elsevier B.V. Summary: The combination of genetically modified maize varieties tolerant to herbicides and resistant to insects, along with the application of glyphosate, may effectively control weeds and pests. However, the impact on nontarget arthropods still needs evaluation. Studying the population changes of canopy-dwelling arthropods in Bt maize under different management options, including glyphosate application, found no significant effects except for the decrease of certain groups in plots with no pre-emergence herbicides, particularly in treatments involving two glyphosate applications.

Copper and kaolin sprays as tools for controlling the olive fruit fly

Summary: The study on controlling the olive fruit fly found that kaolin sprays effectively reduced damage to olives and ensured high-quality oil production. Copper treatments provided some protection but were insufficient at high infestation levels. However, the impact on non-target arthropods was relatively minor.

Summary: Field trials conducted in Central Spain showed that different management strategies for weed and pest control did not have a noticeable impact on the communities of ground-dwelling phytophagous and predatory arthropods, although minor changes were observed in the abundances of some taxa. The use of reduced doses of conventionally used herbicides resulted in increased populations of dominant ground beetle species. The stability of the ground-dwelling arthropod community was not significantly affected by any of the maize varieties or herbicide options assessed in this study.

Summary: The VP60 capsid protein from rabbit haemorrhagic disease virus can form virus-like particles when expressed using different protein expression systems. In this study, a dual-serving RHDV vaccine was successfully developed using CrisBio technology, which allows the co-expression of VP60 proteins from two RHDV serotypes. Rabbits immunised with the chimeric VLPs were fully protected against a lethal challenge infection with the two RHDV serotypes, demonstrating the effectiveness of the dual vaccine strategy.

Development of a Disease and Pest Management Program to Reduce the Use of Pesticides in Sweet-Cherry Orchards

Summary: A new integrated pest management (New IPM) protocol has been implemented in Spain for managing the main diseases and pests of sweet cherry. This protocol includes nonchemical strategies, adjusts the timing and number of pesticide applications based on damage thresholds and a predictive model, and reduces the use of pesticides significantly without affecting the control of diseases and pests.

Evaluation of Mass Trapping Devices for the Control of the European Cherry Fruit Fly [Rhagoletis cerasi (L.)]

Summary: Rhagoletis cerasi (L.) is a damaging pest of cherries in Europe, Western Asia, and North America. The management of this pest mainly relies on insecticide applications near harvest, but the repeated use of insecticides poses environmental risks and may lead to resistance and fruit residues. This study compared trap-attractant combinations and found that a folded yellow sticky trap with a homemade ammonium acetate dispenser was the most efficient and cost-effective device for mass trapping of R. cerasi. Farm-scale trials showed that mass trapping can reduce the need for insecticide applications and keep damage below the economic threshold, making it a useful tool for managing R. cerasi.
