Evaluation of Distinct Freezing Methods and Cryoprotectants for Human Amniotic Fluid Stem Cells Cryopreservation

Amniotic fluid (AF) was described as a potential source of mesenchymal stem cells (MSCs) for biomedicine purposes. Therefore, evaluation of alternative cryoprotectants and freezing protocols capable to maintain the viability and stemness of these cells after cooling is still needed. AF stem cells (AFSCs) were tested for different freezing methods and cryoprotectants. Cell viability, gene expression, surface markers, and plasticity were evaluated after thawing. AFSCs expressed undifferentiated genes Oct4 and Nanog; presented typical markers (CD29, CD44, CD90, and CD105) and were able to differentiate into mesenchymal lineages. All tested cryoprotectants preserved the features of AFSCs however, variations in cell viability were observed. In this concern, dimethyl sulfoxide (Me2SO) showed the best results. The freezing protocols tested did not promote significant changes in the AFSCs viability. Time programmed and nonprogrammed freezing methods could be used for successful AFSCs cryopreservation for 6 months. Although tested cryoprotectants maintained undifferentiated gene expression, typical markers, and plasticity of AFSCs, only Me2SO and glycerol presented workable viability ratios.

Correlation between estimated fetal weight and weight at birth in infants with gastroschisis and omphalocele

Summary: The objective of this study was to evaluate the correlation between estimated fetal weight (EFW) calculated using different formulae and birth weight, as well as to assess the accuracy of classifying fetuses as small for gestational age (SGA) at birth. The results showed a good correlation between EFW and birth weight for infants with gastroschisis, but no correlation for those with omphalocele. All formulae overestimated the number of SGA cases.

Summary: A new technique was standardized to evaluate fetal leg muscle trophism, showing significant differences between normal and fetuses with open lumbosacral spina bifida through measurements of leg area and circumference. The method proved to be highly reproducible, with lower measurements observed in fetuses with SB compared to the control group, and particularly in cases with abnormal lower limb movements.

Hydrocephalus in myelomeningocele

Summary: The study found that the onset of hydrocephalus is directly related to myelomeningocele closure in neurosurgery. In patients who underwent surgery after the first 6 h of life, 83% developed hydrocephalus after birth and required ventriculoperitoneal shunt placement in the maternity ward.

In utero myelomeningocele repair and incidence of lower urinary tract surgery. Results of a prospective study

Summary: The study aimed to determine the incidence of lower urinary tract surgery in patients with myelomeningocele operated in utero, as well as in the subgroup of patients aged 5 years and older. The results showed surgery incidence rates of 10.8% for patients over 5 years old and 12.9% for the high-risk group. This information may be useful to educate parents about the importance of urological treatment.

Ultrasound evaluation of uterine scar thickness after open fetal surgery for myelomeningocele

Summary: This study aimed to analyze the evolution of uterine scar thickness after open fetal surgery for myelomeningocele and establish a cut-off point for uterine scar thickness associated with high risk of uterine rupture. The results showed that thinner uterine scars were significantly associated with increased likelihood of thinning during pregnancy follow-up. A cut-off point of ≤3.0 mm in scar thickness may be useful for decision-making regarding the risk of uterine rupture and indication of caesarean section.

Assessment of lower limb trophism in fetuses with open spina bifida using fractional thigh volume of three-dimensional ultrasound

Summary: This study assessed the trophism of the lower limbs of fetuses with open spina bifida using fractional thigh volume (TVOL) measured by three-dimensional ultrasound. The results showed no significant differences in TVOL between normal fetuses and those with spina bifida, while fetuses with spina bifida and without spontaneous movement of the lower limbs had lower TVOL values.
