The Prominent Role of Neutrophils during the Initial Phase of Infection by Leishmania Parasites

Neutrophils are rapidly and massively recruited to the site of Leishmania inoculation, where they phagocytose the parasites, some of which are able to survive within these first host cells. Neutrophils can thus provide a transient safe shelter for the parasites, prior to their entry into macrophages where they will replicate. In addition, neutrophils release and synthesize rapidly several factors including cytokines and chemokines. The mechanism involved in their rapid recruitment to the site of parasite inoculation, as well as the putative consequences of their massive presence on the microenvironment of the focus of infection will be discussed in the context of the development of the Leishmania-specific immune response.

Summary: Injection of effector proteins by pathogenic organisms to block host innate immune signaling is a common strategy for establishing infection. This study reveals that GSDME is activated in a RIPK1-dependent manner and promotes neutrophil pyroptosis and IL-113 release critical for anti-Yersinia defense, establishing it as an important mediator to counteract pathogen blockade of innate immune signaling.

The c-MET receptor tyrosine kinase contributes to neutrophil-driven pathology in cutaneous leishmaniasis

Summary: This study investigates the role of c-MET receptor tyrosine kinase in the pathology of leishmaniasis. The results show that L. mexicana infection induces the expression and phosphorylation of c-MET in neutrophils. Genetic deficiency of c-MET in neutrophils leads to better control of lesion size and parasite load. In addition, inhibition of c-MET with a specific inhibitor significantly reduces the cutaneous pathology. These findings suggest that c-MET contributes to the development of cutaneous lesions in leishmaniasis, and targeting c-MET may have potential therapeutic value.

Summary: Neutrophils transiently migrate to draining lymph nodes through blood vessels via the C5a-C5aR1 axis, guided by L-selectin and integrins, according to a study using a murine model of dermal infection. The study also found that C5, the C5a precursor, is locally produced in the draining lymph node by lymphatic endothelial cells.
