The Leishmania HSP20 is antigenic during natural infections, but, as DNA vaccine, it does not protect BALB/c mice against experimental L. amazonensis infection

Protozoa of the genus Leishmania are causative agents of leishmaniasis, an important health problem in both human and veterinary medicine. Here, we describe a new heat shock protein (HSP) in Leishmania, belonging to the small HSP ( sHSP) family in kinetoplastids. The protein is highly conserved in different Leishmania species, showing instead significant divergence with sHSP's from other organisms. The humoral response elicited against this protein during Leishmania infection has been investigated in natural infected humans and dogs, and in experimentally infected hamsters. Leishmania HSP20 is a prominent antigen for canine hosts; on the contrary, the protein seems to be a poor antigen for human immune system. Time-course analysis of appearance of anti-HSP20 antibodies in golden hamsters indicated that these antibodies are produced at late stages of the infection, when clinical symptoms of disease are patent. Finally, the protective efficacy of HSP20 was assessed in mice using a DNA vaccine approach prior to challenge with Leishmania amazonensis. Copyright (C) 2008 Ana M. Montalvo-Alvarez et al.

Identification of the interactomes associated with SCD6 and RBP42 proteins in Leishmania braziliensis

Summary: Leishmania parasites exhibit precise gene regulation, mainly at the post-transcriptional level, to adapt to environmental changes. RNA binding proteins SCD6 and RBP42 in L. braziliensis play roles in mRNA-protein complex assembly and gene regulation. The interaction of these proteins with other cellular components could provide new insights for therapeutic targets against Leishmania infections.

Inoculation of the Leishmania infantum HSP70-II Null Mutant Induces Long-Term Protection against L. amazonensis Infection in BALB/c Mice

Summary: The genetically-modified attenuated L. infantum line, Li Delta HSP70-II, was shown to prevent cutaneous leishmaniasis in BALB/c mice challenged with virulent L. amazonensis parasites. Vaccinated mice exhibited decreased disease evolution, reduced lesions, and parasite burdens, along with induction of anti-Leishmania antibodies and specific IFN-gamma responses. This live vaccine may have potential as a candidate for human vaccine development against different forms of leishmaniasis.

Gene Annotation and Transcriptome Delineation on a De Novo Genome Assembly for the Reference Leishmania major Friedlin Strain

Summary: Leishmania major is the main causative agent of cutaneous leishmaniasis in humans. The genome sequence of the Friedlin strain (LmjF) was successfully deciphered in 2005, serving as a reference for subsequent genome assemblies in other Leishmania species. A new assembly for the LMJFC strain was generated using Illumina short-read sequencing and PacBio Single Molecular Real-Time sequencing, improving genome annotation and providing gene models for 11,238 genes.

Live attenuated vaccines, a favorable strategy to provide long-term immunity against protozoan diseases

Summary: Control of diseases caused by protozoan parasites is an important sustainable development goal. Efforts have been made to develop new live attenuated vaccines (LAVs) against malaria, Chagas disease and leishmaniasis. However, challenges related to biosafety, production, and distribution have slowed down further development. Progress has been made in overcoming the drawbacks of LAVs, but the persistence of LAVs and standardized clinical evaluation procedures for vaccine candidates need further clarification and development.

A dataset of proteins associated with Trypanosoma cruzi LYT1 mRNAs

Pull-down assays and proteomic analyses identified proteins that interact with LYT1 mRNA, with some proteins having RNA binding motifs. These findings provide insights into the putative regulatory proteins involved in the processing of LYT1 mRNAs.

Assembly of a Large Collection of Maxicircle Sequences and Their Usefulness for Leishmania Taxonomy and Strain Typing

Summary: This study assembled the complete coding region of maxicircles for different trypanosomatid species based on various databases and a bioinformatics workflow. The phylogenetic analysis using the assembled data showed accurate taxonomy and discrimination of closely related Leishmania species. Additionally, a dataset of maxicircle sequences for L. infantum field isolates was provided, revealing the existence of different populations/clades within the Mediterranean region and establishing maxicircle signatures for each clade.

Neglected Zoonotic Diseases: Advances in the Development of Cell-Penetrating and Antimicrobial Peptides against Leishmaniosis and Chagas Disease

Summary: In 2020, the WHO established a road map for neglected tropical diseases 2021-2030, aiming to control and eradicate 20 diseases including leishmaniosis and Chagas disease. The WHO has also been developing a Global Action Plan on Antimicrobial Resistance since 2015. Natural peptide molecules (AMPs and CPPs) show promise as alternative therapies for neglected zoonoses by improving efficacy while avoiding toxicity and resistance. Encapsulation and functionalization methods are being used to overcome limitations of antimicrobial peptides for approval in clinical programs against leishmaniosis and Chagas disease.
