Summary: This study investigated the biological effects and osteoinductivity of the combination of demineralized dentin matrix (DDM) and fibrin glue (FG). In vitro studies showed that a ratio of 1:0.1 of DDM to FG induced more alkaline phosphatase (AIP) activity and mineralized nodule formation, while a ratio of 1:0.5 induced more osteocalcin (OC) production. In an animal model, the DDM-FG compound enhanced osteogenic activities and bone regeneration, with DDM-FG2 showing greater new bone volume during the early healing period.

The results showed that the use of the HA-DBM scaffold led to significantly less soft tissue edema at the fusion site compared to rhBMP-2 treatment, with no elevation in certain cytokines observed in the HA-DBM group.

Summary: A highly bioactive mesoporous glass solvBG76 was successfully synthesized using a facile method. The material showed nano-size, non-spherical morphology, and a structure free of crystalline impurities, enhancing the deposition of hydroxyapatite (HA) when in contact with physiological fluids. Various analyses confirmed the formation of HA layer on the glass, and solvBG76 exhibited superior apatite-mineralization ability compared to other glasses. Moreover, it demonstrated biocompatibility and hemocompatibility.

Summary: This study compared the osteoinductive potential of BMP-2 ho-modimer with a heterodimer of BMP-2/7 in a collagen-hydroxyapatite (CHA) scaffold delivery system. The results showed that CHA + BMP-2/7 had higher osteoinductive ability than CHA + BMP-2, and the CHA scaffold provided sustained release of BMP-2/7 for at least 20 days. Additionally, a low dose of BMP-2/7 accelerated fracture healing and performed at a level comparable to a 20-times higher BMP-2 dose, bringing hope for the use of physiological growth factor doses in fracture healing.

Summary: In this study, mineralized collagen (MC) was incorporated into polymethylmethacrylate (PMMA) bone cement to enhance osteointegration ability, showing improved wettability and mechanical performance of PMMA-MC in vitro. In animal models, the PMMA-MC group demonstrated more new bone formation and higher fixation force compared to the PMMA group. Moreover, in torsion tests on cadaveric femurs, the PMMA-MC group required more torque for the same rotation degree, indicating the potential of MC to induce efficient fixation in osteoporotic bone.

Summary: Bone morphogenetic proteins (BMPs) require a suitable delivery system for treating spinal and bone defects. INFUSE, the only commercially available BMP-based device, has major disadvantages. A novel device called OSTEOGROW, containing BMP6 dispersed within autologous blood coagulum, shows promising results for bone regeneration. This study compares the release dynamics and cellular response of OSTEOGROW and INFUSE implants and demonstrates that OSTEOGROW is superior in inducing bone formation.
