Summary: Studies have shown that culturing MSCs in a low oxygen environment can upregulate stem cell markers and promote cell proliferation in an undifferentiated state. However, there is ambiguity in the results due to differences in oxygen gradients and culture conditions among different studies. This progress report focuses on conflicting findings related to the application of hypoxic conditions for improving MSC proliferation or differentiation, aiming to decipher the factors influencing MSC characteristics under hypoxia.

Summary: This study investigated the impact of osteogenic differentiation on the immunomodulatory properties of hPDL-MSCs, and found that osteogenic differentiation reduced the ability of hPDL-MSCs to suppress CD4(+) T lymphocyte proliferation and enhanced this ability in the presence of IL-1 beta. Additionally, the expression and activity of indoleamine-2,3-dioxygenase and interleukin-8 were also affected by the differentiation process.

Summary: A method based on single-cell FTIR microspectroscopy was developed to quantitatively evaluate cellular heterogeneity by calculating the cell-to-cell similarity distance, revealing that infrared phenotypes may reflect dynamic heterogeneity changes in cell populations. This provides an alternative label-free optical approach for quantifying cellular heterogeneity and can be combined with other single-cell analysis tools to understand the genotype-to-phenotype relationship in cellular populations.

Summary: In the latest research, mesenchymal stem cells derived exosomes (MSC-Exos) are identified as a potential alternative for treating refractory diabetic wounds. They have similar biological activity but less immunogenicity compared to MSCs. This review focuses on summarizing the effects of different MSC-Exos on diabetic wounds based on their origins and contents, discussing experimental conditions, target cells/pathways, and mechanisms. Additionally, the combination of MSC-Exos and biomaterials is explored to improve therapy efficacy and utilization. The clinical value and application prospects of exosome therapy, as well as the development trends of using exosomes as carriers for targeted delivery, are highlighted.

Summary: This study compared the bioactivity of free fucoidan and fucoidan-loaded nanoparticles in human hepatoma cells. The results showed that nanoparticles reduced cell invasion and migration, while free fucoidan increased these events at low concentrations. Moreover, nanoparticles decreased the production of reactive oxygen species and the activity of matrix metalloproteinase-9 compared to free fucoidan. Proteomic analysis revealed that fucoidan regulated proteins associated with protein synthesis, N-glycan processing, and cellular stress.

Summary: The invasive species Rugulopteryx okamurae, a brown macroalgae, has been found in European waters and the Strait of Gibraltar. Its proliferation poses significant threats to local ecosystems and results in a socioeconomic burden due to the accumulation of biomass as waste. Researchers have explored using the algae biomass in value-added products or technologies to minimize its impact. This review highlights the potential uses of R. okamurae, such as biogas production, composting, bioplastic, and pharmaceutical purposes.

Summary: Diabetic foot ulcers (DFUs) are complex and severe complications of diabetes, often exacerbated by inflammation and infections. Current dressing systems combined with antibiotic or immunomodulatory therapies have limitations in fighting microbial presence and providing a suitable environment for wound healing. Research in this field has rapidly expanded in recent years to develop new and more effective therapeutic systems for DFU care. This review compiles emerging innovations in DFU dressing systems using fiber-based scaffolds modified with bioactive compounds, focusing on advancements from 2017 to 2022.

Summary: In chronic wound (CW) scenarios, Staphylococcus aureus-induced infections are very prevalent. This leads to abnormal inflammatory processes, in which proteolytic enzymes, such as human neutrophil elastase (HNE), become highly expressed. A co-axial drug delivery system was developed to incorporate the antimicrobial peptide AAPV, which can suppress HNE activity, and release it controlled by N-carboxymethyl chitosan (NCMC), an antimicrobial polymer effective against Staphylococcus aureus.

Summary: Wound infection is a major obstacle to adequate healing, especially in burn wounds and chronic wounds. Silver nanoparticles (AgNPs) are effective antimicrobial agents used in wound dressings, but their cytotoxicity and potential neurotoxic effects are concerns. This study proposes two strategies to immobilize and increase the concentration of AgNPs in wound dressings to avoid systemic cytotoxicity and enhance their antimicrobial properties. The effectiveness of the composite materials against bacteria and a bacteriophage was tested, providing insights into their antimicrobial capabilities and the release of AgNPs.

Summary: Infection is a major issue in chronic wound care. This research aims to find an effective solution for preventing microbial propagation in wound dressing. Polyvinyl alcohol nanofibrous mats reinforced with cellulose nanocrystal, crosslinked with glutaraldehyde vapor, and doped with specialized peptides were produced. Antimicrobial assessments demonstrated the effectiveness of the peptide-doped mats against Staphylococcus aureus and Pseudomonas aeruginosa. C90/10 PVA/CNC mats were deemed the most effective option.
