Carbohydrate Sequence of the Prostate Cancer-associated Antigen F77 Assigned by a Mucin O-Glycome Designer Array

Monoclonal antibody F77 was previously raised against human prostate cancer cells and has been shown to recognize a carbohydrate antigen, but the carbohydrate sequence of the antigen was elusive. Here, we make multifaceted approaches to characterize F77 antigen, including binding analyses with the glycolipid extract of the prostate cancer cell line PC3, microarrays with sequence-defined glycan probes, and designer arrays from the O-glycome of an antigen-positive mucin, in conjunction with mass spectrometry. Our results reveal F77 antigen to be expressed on blood group H on a 6-linked branch of a poly-N-acetyllactosamine backbone. We show that mAb F77 can also bind to blood group A and B analogs but with lower intensities. We propose that the close association of F77 antigen with prostate cancers is a consequence of increased blood group H expression together with up-regulated branching enzymes. This is in contrast to other epithelial cancers that have up-regulated branching enzymes but diminished expression of H antigen. With knowledge of the structure and prevalence of F77 antigen in prostate cancer, the way is open to explore rationally its application as a biomarker to detect F77-positive circulating prostate cancer-derived glycoproteins and tumor cells.

GIPS-Mix for Accurate Identification of Isomeric Components in Glycan Mixtures Using Intelligent Group-Opting Strategy

Summary: Accurate identification of glycan structures is challenging due to the existence of multiple isomeric structures in glycan samples. Researchers developed an intelligent group-opting strategy, named GIPS-mix, to assign isomeric glycans within mixtures based on simulated spectra and experimental spectra similarity. If a single group cannot be elected, a tie break is performed by additional MSn scanning using intelligently selected precursors. Through the application of GIPS-mix on standard mixtures and human milk oligosaccharide fractions, high accuracy and efficiency in assigning individual glycans in mixtures were demonstrated.

Summary: Brewing practice involves reusing the same yeast for subsequent fermentations. The yeast used for Lager beer (Saccharomyces pastorianus) is widely reused without changes in fermentation performance. However, the yeast used for Ale beer (Saccharomyces cerevisiae) is either partially or not reused due to poor performance. The study found that cells modulate their wall polysaccharides to increase cell wall strength.

Feasibility of Brewer's Spent Yeast Microcapsules as Targeted Oral Carriers

Summary: This study aims to investigate the feasibility of brewer's spent yeast (BSY) microcapsules as oral carriers for food and biomedical applications. The results demonstrate that BSY microcapsules have strong resistance to digestion and are rich in (beta 1 -> 3)-glucans, which can be recognized by the human Dectin-1 immune receptor. Additionally, the digested material is recognized by various lectins related to (beta 1 -> 3)-glucans, glycogen, and mannans.

Dual kinase inhibitor for EGFR mutants and ErbB2 limit breast cancer

Summary: Mutations in the EGFR gene are common in NSCLC, with the L858R mutation in Exon 21 being the most prevalent. While targeted therapies like monoclonal antibodies and TKIs have shown efficacy, resistance eventually develops due to mutations such as T790M and C797S. A third-generation TKI, AZD9291, has high affinity for these mutations, but recent research suggests the emergence of resistance due to the EGFR (C797S) mutation. Additionally, erbB2 amplification is another significant mechanism of resistance. A small kinase inhibitor, ER121, has been developed to target these mutations and has shown promising inhibitory activity in mutant EGFR and amplified ErbB2 cancers.

Structural and functional analysis of natural capsid variants suggests sialic acid-independent entry of BK polyomavirus

Summary: This study characterizes the impact of naturally occurring VP1 mutations on ganglioside binding, VP1 protein structure, and virus tropism in BK polyomavirus (BKPyV). Mutants E73Q, E73A, and A72V-E73Q-E82Q (VQQ) retain sialic acid-dependent infectious entry and bind to a-series gangliosides, including GD1a. However, E73A and VQQ mutants lose the ability to infect ganglioside-complemented cells, and this correlates with a BC2 loop shift in their crystal structures. The K69N mutation in the K69N-E82Q variant inhibits sialic acid binding but retains significant infectivity in 293TT cells, suggesting the existence of an unknown sialic acid-independent entry receptor for BKPyV.

However, there is limited research on the specific differences in HMOs as a function of gestation time. In this study, we established an approach to analyze HMOs and found that the composition and relative abundance of HMOs changed with postpartum age, with significant differences in HMOs secreted at different postpartum times.
