Up-regulation of miR-5581 inhibited cell proliferation, extracellular matrix (ECM) synthesis, and promoted ECM degradation by targeting NRF1, where Kras signaling might be involved. NRF1 was down-regulated in knee joint cartilage of OA mice.

Summary: This experiment investigates the role of Sox9 in cardiomyocyte apoptosis after acute myocardial infarction (AMI). The expression levels of Sox9, miR-223-3p, and MEF2C in cardiomyocyte apoptosis are studied, and their interactions are verified. The results show that high expression of Sox9 promotes cardiomyocyte apoptosis, while silencing of Sox9 protects cardiomyocytes from apoptosis by enhancing cell viability, suppressing apoptosis, and reducing caspase-3 and caspase-9 activity. The experiment also reveals that Sox9 exacerbates hypoxia-induced cardiomyocyte apoptosis by upregulating miR-223-3p expression and inhibiting MEF2C transcription.

Summary: Upregulation of miR-181a promotes the development of cutaneous squamous cell carcinoma (cSCC) by targeting TGF beta R3, a component of TGF beta signaling. Overexpression of miR-181a or knockdown of TGF beta R3 significantly suppresses UV-induced apoptosis in HaCaT cells and normal human epidermal keratinocytes. Moreover, miR-181a overexpression or TGF beta R3 knockdown enhances the survival, migration, and invasion of tumor cells.

Summary: Selective loss of Tcf7l2 gene expression in adipocytes leads to a sexually dimorphic phenotype, with impairments not only in adipocytes, but also in pancreatic islet and enteroendocrine cells in male mice only. Our findings suggest novel roles for adipokines and incretins in the effects of diabetes-associated variants in TCF7L2, and further illuminate the roles of TCF7L2 in glucose homeostasis and diabetes risk.

Summary: This study demonstrates that the elevated expression of miR-125b-5p is associated with impaired pancreatic beta-cell function in type 2 diabetes, and overexpression of this miRNA impairs glucose-stimulated insulin secretion. Screening identifies multiple targets of miR-125b-5p, including a transporter and a regulator of organelle dynamics. In human beta cells, silencing miR-125b-5p enhances insulin secretion, while overexpression in a mouse model leads to hyperglycemia and glucose intolerance.
