Summary: Combination therapy with PD-1 blockade and IL-2 improves anti-tumor efficacy, but the underlying mechanisms are not well understood. Our study reveals that PD-1 blockade enhances IL-2 effect by regulating the MARCH5 and γc signaling pathways, leading to potential benefits in cancer immunotherapy.

Summary: This study investigated the regulatory function of WISP1 in kidney inflammation. The results showed that WISP1 can induce kidney inflammation and it can be prevented by inhibiting NF-KB. In addition, inhibition of WISP1 can suppress the production of inflammatory cytokines in macrophages and the proliferation of kidney fibroblasts. The increased expression of WISP1 in kidney inflammation models was also confirmed. Therefore, pharmacological blockade of WISP1 shows potential as a novel therapy for kidney inflammation.

Summary: This study reveals that the plasma membrane-associated E3 ubiquitin ligase MARCH3 negatively regulates IL-3-triggered signaling. MARCH3 is associated with IL-3R alpha, promoting its degradation and inhibiting IL-3-induced physiological functions and inflammatory diseases. MARCH3 deficiency aggravates IL-3-amplified expression of inflammatory cytokines, organ damage, and inflammatory death in sepsis.

Summary: This study reveals that Scalloped, as a downstream transcription factor of the Hippo signaling pathway, is upregulated in shrimp hemocytes and interacts with the Hippo pathway downstream transcriptional co-activator Yki. By inhibiting the Dorsal pathway, Scalloped reduces the expression of immune functional proteins and negatively regulates the immune response against bacterial infection in shrimp.

Summary: Forskolin attenuates cisplatin-induced auditory cell death by activating cAMP/PKA/CREB signaling, with potentiating effects when combined with gap junction enhancers. This study provides insight into the prevention of cisplatin-induced ototoxicity through gap junction-mediated intercellular communication of cAMP.

Summary: This study investigates the mechanism of dbcAMP on porcine fat deposition. The results suggest that dbcAMP regulates gene expression related to adipocyte differentiation and fat metabolism through the cAMP-PKA pathway, leading to reduced fat deposition.

Summary: The study found that phosphorylation of the transcription factor CREB correlates strongly with survival in medulloblastoma patients and differentiation phenotype. During normal cerebellar development, phosphorylated CREB is selectively expressed in differentiating cerebellar granule neuron progenitor cells. Inducing CREB activation via PKA-mediated signaling enhances sensitivity of medulloblastoma cells to chemotherapy.

Summary: Nervous system malignancies exhibit rapid progression and low survival rates. NR5A2, identified as a negative regulator of cancer cell proliferation, shows potential as a pharmacological target for nervous system-related tumors. High expression levels of NR5A2 are associated with a favorable prognosis in patients with glioblastoma and neuroblastoma, with the antiproliferative effect of NR5A2 mediated by the induction of negative regulators of the cell cycle. Agonists of NR5A2, such as DLPC, mimic its antiproliferative action in glioblastoma cells and inhibit tumor growth in mouse models. These findings suggest a tumor suppressor role of NR5A2 in the nervous system, highlighting its potential as a therapeutic target for nervous tissue-related tumors.
