Cell fate and proliferation are tightly linked to the regulation of the mitochondrial energy metabolism. Hence, mitochondrial biogenesis regulation, a complex process that requires a tight coordination in the expression of the nuclear and mitochondrial genomes, has a major impact on cell fate and is of high importance. Here, we studied the molecular mechanisms involved in the regulation of mitochondrial biogenesis through a nutrient-sensing pathway, the Ras-cAMP pathway. Activation of this pathway induces a decrease in the cellular phosphate potential that alleviates the redox pressure on the mitochondrial respiratory chain. One of the cellular consequences of this modulation of cellular phosphate potential is an increase in the cellular glutathione redox state. The redox state of the glutathione disulfide-glutathione couple is a well known important indicator of the cellular redox environment, which is itself tightly linked to mitochondrial activity, mitochondria being the main cellular producer of reactive oxygen species. The master regulator of mitochondrial biogenesis in yeast (i.e. the transcriptional co-activator Hap4p) is positively regulated by the cellular glutathione redox state. Using a strain that is unable to modulate its glutathione redox state (Delta glr1), we pinpoint a positive feedback loop between this redox state and the control of mitochondrial biogenesis. This is the first time that control of mitochondrial biogenesis through glutathione redox state has been shown.

The Dct-/- Mouse Model to Unravel Retinogenesis Misregulation in Patients with Albinism

Summary: We have identified DCT encoding dopachrome tautomerase (DCT) as the eighth gene for oculocutaneous albinism. Loss of DCT function leads to eye hypopigmentation and retinal dystrophy. By investigating Dct mice, we found severe hypopigmentation in their retinal pigmented epithelium (RPE) from early stages, with specific cellular defects. Understanding the molecular regulation of retinal development and aging of the hypopigmented eye may help guide therapeutic strategies for patients with albinism.

Summary: This study investigated the differential impact of resveratrol on tumor cells and healthy cells by evaluating its metabolism and the expression of UGT1A genes. The findings revealed that resveratrol was more cytotoxic to tumor cells with downregulated UGTs, while overexpression of UGT1A10 gene reduced its cytotoxicity. Resveratrol also affected the cell cycle, nucleotide metabolism, and global transcription and translation in intestinal sensitive tumor cells. This study enhances our understanding of the antitumor action of resveratrol and provides insights for its selective treatment in colorectal tumors.

Mechanisms of systemic low-grade inflammation in HIV patients on long-term suppressive antiretroviral therapy: the inflammasome hypothesis

Summary: The objective of this study was to determine the contribution of inflammasome activation in chronic low-grade systemic inflammation observed in patients with HIV on long-term suppressive antiretroviral therapy. Inflammasome activation was measured by cytokine analysis in patient serum and ex vivo stimulation of primary monocytes. The results showed higher levels of IL-18 in HIV-positive patients compared to HIV-negative controls, along with increased secretion of IL-1 beta in response to inflammasome stimulation. Oxidative stress was also observed in patient sera. In conclusion, HIV infection with long-term effective ART is associated with inflammasome activation and increased monocyte activation, possibly mediated by oxidative stress.

Summary: This study investigates the developmental requirements for purine metabolism using the Xenopus laevis model. The research shows that purine pathway genes are mainly expressed in nervous and muscular embryonic tissues and are essential for myogenesis. Knockdown of certain genes in the pathway results in defects in muscle formation.
