Summary: Liver fibrosis is a progressive and debilitating disease characterized by excessive deposition of extracellular matrix proteins. Our study reveals that KLF10 acts as a negative regulator of TGF-beta signaling pathway, suppressing hepatic stellate cell activation and fibrogenesis through modulation of ATF3 expression. Targeting the KLF10-ATF3 axis may have therapeutic implications for liver fibrosis treatment.

Summary: This study identifies high YWHAZ and NDRG1 expression as molecular biomarkers associated with poor prognosis in prostate cancer patients, defining two subgroups with high and intermediate risk of disease progression. YWHAZ outperforms Gleason score in predicting aggressiveness of prostate cancer beyond 60 months post-diagnosis.

Summary: Through a chemical genetics screen and phospho-proteomics, Ledet and Ruff et al identify substrates of the oncogenic kinase PIM1 in prostate cancer cells. They find that NDRG1 protein is destabilized by PIM1-mediated phosphorylation to reduce its metastasis suppressor function. These findings shed light on the role of PIM1 in prostate cancer.

Summary: The study found that TRPM4 is a novel methylated tumor suppressor gene that is downregulated in colorectal cancer. TRPM4 activates calpain-mediated cleavage of FAK through modulating the PI3K/Akt/mTOR signaling cascade, thereby impeding CRC migration and invasion.

Summary: This study found that overexpression of miR-7 can suppress tumor growth and prolong survival in transgenic mouse models of prostate cancer. miR-7 inhibits glycolysis in prostate cancer cells by regulating multiple key pathways and remodels the acidic tumor microenvironment. These findings provide hope for using miR-7 as a target for treating prostate cancer.

Summary: Metastatic progression is a key feature of prostate cancer, with RAD9 being an oncogene involved in regulating genes critical for prostate carcinogenesis. Knocking down RAD9 expression affects cell activities and downstream genes involved in prostate cancer. Ectopic expression of FOXP1 or NDRG1 can partially restore RAD9-regulated activities in prostate cancer cells.

Summary: The study revealed that in gastric cancer patients, the down-regulation of NDRG1 expression was negatively correlated with DNMT1, DNMT3A, and DNMT3B, while the DNA methylation of NDRG1 was positively correlated with the DNMT family. High expression of NDRG1 may be associated with prolonged overall survival.

Summary: Colorectal cancer is a common and deadly disease. Protein Kinase C Delta (PKC delta) has been found to play a crucial role in promoting the invasion and migration of cancer cells in colorectal cancer. This is achieved by inhibiting the expression of N-myc Downstream Regulated Gene 1 (NDRG1).

Summary: Samd9/9L syndromes are a group of inherited bone marrow failure syndromes that are caused by gain-of-function mutations in the genes located on chromosome 7. These syndromes often lead to infantile myelodysplastic syndromes associated with monosomy 7, characterized by bone marrow failure and multiorgan degeneration.

Summary: This study confirmed the direct effects of BCAA on human astrocytes, showing negative impacts on cell survival and intracellular metabolism. BCAA treatment led to increased leakage of lactate dehydrogenase and decreased cell proliferation rate, affecting glycolysis and amino acid metabolism within the cells. Furthermore, BCAA treatment was found to decrease levels of Glut-1 and phosphorylated GSK-3 beta and mTOR in astrocytes.

Summary: Gut microbiota play important roles in maintaining homeostasis, regulating immunity, and barrier function in the intestine, among others. Dysbiosis, the disruption in microbiota, is associated with various chronic inflammatory conditions, food allergies, and colorectal cancer. The gut microbiota is influenced by factors such as diet, antibiotics, medications, and bacterial and viral infections. Additionally, the disruption of the oral-gut-liver axis has been linked to non-alcoholic fatty liver disease (NAFLD), the most common liver disease in developed countries.
