Summary: The study revealed that suspension-cultured cells of bamboo produce 3-O-feruloylquinic acid and 3-O-p-coumaroylquinic acid after treatment with SBHA, which is attributed to the action of HQT. A new HQT, named BmHQT1, was identified in this study, and it catalyzes the formation of 3-O-hydroxycinnamoylquinic acid esters.

Summary: The membrane-bound O-acyltransferase (MBOAT) superfamily catalyses the transfer of acyl chains to substrates implicated in essential cellular functions. Aberrant function of MBOATs is associated with various diseases and MBOATs are promising drug targets. Recent progress in structural characterisation of MBOATs has advanced our understanding of their functional mechanism. Integration of information across the MBOAT family has identified a common MBOAT fold and provided insights into substrate and inhibitor engagement. Further studies should focus on characterising MBOATs within their membrane environment as lipid-associated proteins.

Summary: Wnt gradients trigger diverse cellular responses, and PORCN plays a crucial role in modifying Wnt proteins. Despite the unclear structure of PORCN, a new model of its membrane topology has been proposed, which could facilitate the understanding of its biological activity and potential therapeutic targets.

Summary: Natural products, such as antibiotics, anticancer therapeutics, and biofuels, are essential for various applications. Polyketides, a class of structurally diverse secondary metabolites, are synthesized by polyketide synthases (PKSs). While PKS genes are found across different organisms, the ones from eukaryotic organisms are understudied. In this study, a type I PKS from the eukaryotic parasite Toxoplasma gondii, TgPKS2, was characterized through genome mining and biochemical characterization. The ACP domains within TgPKS2 were found to have distinct properties, including self-acylation without the need for an AT domain. This research expands our understanding of biosynthetic enzymes from eukaryotes.

Summary: S-acylation is an important posttranslational modification that regulates cellular processes. This reversible lipid modification affects cellular pathways and physiological processes, and the enzymes and proteins involved in S-acylation are still being discovered and studied.

Summary: Two Phase I studies showed that BI 1356225, a GOAT inhibitor, was well-tolerated in healthy males and subjects with overweight/obesity. However, treatment with BI 1356225 for 28 days did not lead to significant reduction in bodyweight or changes in appetite, eating control, or energy intake.

Summary: This study found that Mycobacterium tuberculosis (Mtb) infection can increase the expression of guanylate-binding proteins (GBPs). Inhibiting GBPs can reduce the release of inflammatory cytokines and suppress Mtb intracellular survival. The methylation status of the GBP1 promoter does not change significantly upon Mtb infection.

Summary: This study found that 1-O-Acylceramides (1-OACs) are natural components of human and mouse epidermis, and are formed shortly before birth in mice. Human keratinocytes are one source of epidermal 1-OACs. The levels of 1-OACs are correlated with deficient skin barrier function in mutant mice with deficiencies of ceramide synthases and diacylglycerol acyltransferases. The synthesis of 1-OACs involves multiple enzymes or an unknown acyltransferase of the epidermis.

Summary: The STC[D/Y] motif in AIP-I/IV has a unique structural or chemical motive to avoid S-to-O acyl transfer, retaining its CTP form intact. Substitution of Gly for the inner Asp/Tyr or Met residues drastically alters the resistance to S-to-O acyl transfer, leading to the formation of CDP products.
