New Insights into the Mechanisms whereby Low Molecular Weight CCR5 Ligands Inhibit HIV-1 Infection

CC chemokine receptor 5 (CCR5) is a G-protein-coupled receptor for the chemokines CCL3, -4, and -5 and a coreceptor for entry of R5-tropic strains of human immunodeficiency virus type 1 (HIV-1) into CD4(+) T-cells. We investigated the mechanisms whereby nonpeptidic, low molecular weight CCR5 ligands block HIV-1 entry and infection. Displacement binding assays and dissociation kinetics demonstrated that two of these molecules, i.e. TAK779 and maraviroc (MVC), inhibit CCL3 and the HIV-1 envelope glycoprotein gp120 binding to CCR5 by a noncompetitive and allosteric mechanism, supporting the view that they bind to regions of CCR5 distinct from the gp120- and CCL3-binding sites. We observed that TAK779 and MVC are full and weak inverse agonists for CCR5, respectively, indicating that they stabilize distinct CCR5 conformations with impaired abilities to activate G-proteins. Dissociation of [I-125] CCL3 from CCR5 was accelerated by TAK779, to a lesser extent by MVC, and by GTP analogs, suggesting that inverse agonism contributes to allosteric inhibition of the chemokine binding to CCR5. TAK779 and MVC also promote dissociation of [S-35] gp120 from CCR5 with an efficiency that correlates with their ability to act as inverse agonists. Displacement experiments revealed that affinities of MVC and TAK779 for the [S-35]gp120-binding receptors are in the same range (IC50 similar to 6.4 versus 22 nM), although we found that MVC is 100-fold more potent than TAK779 for inhibiting HIV infection. This suggests that allosteric CCR5 inhibitors not only act by blocking gp120 binding but also alter distinct steps of CCR5 usage in the course of HIV infection.

Summary: This study shows that immunization with new envelopes (VLP formulated) from individuals with early broadly neutralizing activity induced specific HIV-1 antibodies in rabbits after a short immunization protocol. Summary: Our study shows that individuals recovered from COVID-19 only need one vaccine shot to generate high levels of immune responses, while naive subjects require two doses. However, both groups show comparable neutralizing antibodies and S-specific B cell levels in the late post-vaccination period. In terms of cellular responses, naive individuals exhibit higher levels of immune cell activation and proliferation compared to individuals recovered from COVID-19 in the early post-vaccination period, but after almost 8 months post-vaccination, both groups have comparable cellular responses.

Summary: We developed a rapid platform for studying SARS-CoV-2 by using primary human lung tissue cells directly, which can identify viral targets, express viral entry factors, and screen viral entry inhibitors and anti-inflammatory compounds. The method preserves main immune and structural cell populations, including the most susceptible cell targets for SARS-CoV-2, and maintains the expression of proteins involved in viral infection. Additionally, the method allows for highly reproducible antiviral testing and provides the identification of new compounds missed by conventional systems like VeroE6.

One class classification for the detection of β2 adrenergic receptor agonists using single-ligand dynamic interaction data

Summary: G protein-coupled receptors play a crucial role in relaying extracellular signals inside the cell. This study proposes a new method based on the conformational dynamics of a protein-ligand complex to improve the selection of ligands with specific pharmacological properties in structure-based virtual screening. Molecular dynamics simulations were used to extract interaction patterns between an agonist and the beta 2 adrenergic receptor, which were then encoded in graphs and used to train a machine learning classifier. The results demonstrate that the consistency of the binding mode during the simulation is important for the success of the method.

Treatment with integrase inhibitors alters SARS-CoV-2 neutralization levels measured with HIV-based pseudotypes in people living with HIV

Summary: This study compares two methods using HIV and VSV-based pseudoviruses to detect neutralizing antibodies (NAbs) against SARS-CoV-2 in people living with HIV (PLWH). The HIV-pseudoparticles method shows higher neutralization titers, but also detects NAbs in COVID-19-naive PLWH. Treatment with integrase inhibitors can lead to inaccurate results when using the HIV-based pseudoviruses method, but this interference can be eliminated through IgG purification.

Summary: The humoral immune response against SARS-CoV-2 variants of concern after vaccination was evaluated in COVID-19 recovered individuals. It was found that recovered individuals had higher levels of antibodies after a single dose compared to naive participants after two doses. Furthermore, antibodies against the receptor-binding domain were higher in individuals who had recovered for 4-12 months compared to those who had recovered for 1-3 months, and the neutralizing potency against variants of concern was also higher in the longer recovered group.

Summary: Tyrosine kinase inhibitors (TKIs) are widely used to treat chronic myeloid leukemia (CML). Dasatinib, a broad-spectrum TKI with off-target effects, has immunomodulatory capabilities that enhance innate immune responses against cancerous and viral-infected cells. Studies have shown that dasatinib expands memory-like natural killer (NK) cells and γδ T cells, which are associated with improved control of CML and HIV infection. Additionally, dasatinib has potential as a senolytic drug by inducing apoptosis in senescent cells. This review examines the virological and immunogenetic factors associated with dasatinib's cytotoxic effects and discusses its therapeutic potential against CML, HIV infection, and aging.
