By scaffolding MAPK signaling components, beta-arrestins initiate a second wave of signaling, promoting the activation of ERK1/2 or JNK3. Understanding the molecular and structural mechanisms of beta-arrestin-mediated MAPK scaffolding assembly is crucial for understanding GPCR-mediated MAPK activation and selectively regulating the MAPK signaling cascade for therapeutic purposes.

There is no approved pharmacotherapy for MA use disorder, and little is known about the neurobiological determinants of vulnerability to the disease. Studies suggest that ERK activation in the medial prefrontal cortex may play a role in the affective valence of MA and its reinforcing properties.

Two different modes of endocytic activity have been discovered, one dependent on the C-terminus and the other dependent on the C-lobe of beta-arrestins. Summary: This study investigated the impact of phosphorylation modifications in a specific sequence of the CB1 receptor C-terminus on the translocation of Arrestin-2 and Arrestin-3. The results showed that these modifications partially contributed to the translocation of Arrestin, but complete inhibition was only achieved when all phosphorylation sites were mutated. Additionally, the dissociation of G proteins was also impaired proportionally to the extent of Arrestin translocation.
