Protection of Neurons from Apoptosis by Apolipoprotein E-containing Lipoproteins Does Not Require Lipoprotein Uptake and Involves Activation of Phospholipase Cγ1 and Inhibition of Calcineurin

Apolipoprotein E-containing lipoproteins (LpE) are generated in the central nervous system by glial cells, primarily astrocytes, and are recognized as key players in lipid metabolism and transport in the brain. We previously reported that LpE protect retinal ganglion neurons from apoptosis induced by withdrawal of trophic additives (Hayashi, H., Campenot, R. B., Vance, D. E., and Vance, J. E. (2007) J. Neurosci. 27, 1933-1941). LpE bind to low density lipoprotein receptor-related protein-1 and initiate a signaling pathway that involves activation of protein kinase C delta and inhibition of the pro-apoptotic glycogen synthase kinase 3 beta. We now show that uptake of LpE is not required for the neuroprotection. Experiments with inhibitors of phospholipase C gamma 1 and RNAi knockdown studies demonstrate that activation of phospholipase C gamma 1 is required for the anti-apoptotic signaling pathway induced by LpE. In addition, the protein phosphatase-2B, calcineurin, is involved in a neuronal death pathway induced by removal of trophic additives, and LpE inhibit calcineurin activation. LpE also attenuate neuronal death caused by oxidative stress. Moreover, physiologically relevant apoE3-containing lipoproteins generated by apoE3 knock-in mouse astrocytes more effectively protect neurons from apoptosis than do apoE4-containing lipoproteins. Because inheritance of the apoE4 allele is the strongest known genetic risk factor for Alzheimer disease, the reduced neuroprotection afforded by apoE4-containing LpE might contribute to the neurodegeneration characteristic of this disease.

Effects of laminin-111 peptide coatings on rat neural stem/progenitor cell culture

Cytotoxic Effects of Arsenite in Combination With Gamabufotalin Against Human Glioblastoma Cell Lines

Summary: Glioblastoma is a fatal primary malignant brain tumor with a low survival rate, and there is an urgent need for novel therapeutic approaches. Trivalent arsenic derivative and gamabufotalin show cytotoxic effects against glioblastoma cells, with synergistic cytotoxicity when combined. The study suggests that G(2)/M cell cycle arrest, necrosis, and autophagy contribute to the cytotoxicity, and a possible strategy of combining these compounds with a p38 MAPK inhibitor may provide new insights into combating glioblastoma.

Effect of Progranulin on Proliferation and Differentiation of Neural Stem/Progenitor Cells after Oxygen/Glucose Deprivation

Summary: In this study, the researchers investigated the effects of sivelestat and progranulin (PGRN) on neural stem cells under ischemic conditions. They found that treatment with recombinant PGRN (rPGRN) increased cell proliferation and phosphorylation of Akt and GSK-3 beta, while sivelestat had no effect. Additionally, rPGRN enhanced neurite outgrowth of differentiated neurons. These findings suggest that PGRN could be a potential therapeutic approach for cerebral ischemia.

Possible Involvement of DNA Methylation and Protective Effect of Zebularine on Neuronal Cell Death after Glutamate Excitotoxity

Summary: Neuronal cell death after cerebral ischemia is mainly caused by excessive Ca2+ influx through NMDA receptors. DNA methylation plays an important role in this process, and inhibition of DNMTs can protect cells from injury. Pharmacological inhibition of DNMTs is effective in protecting against glutamate excitotoxicity.

Summary: The study found that E-LPs can protect the optic nerve from degeneration and reduce the increase of alpha-2-macroglobulin in the aqueous humor and retina of rats. E-LPs directly decreased the expression and secretion of alpha-2-macroglobulin in primary cultures of Muller glia, which was blocked by knockdown of LRP1. E-LPs promoted the phosphorylation of STAT3 and inhibited the expression of alpha-2-macroglobulin, suggesting a potential protective mechanism against excitotoxicity in the retina.
