Summary: The human pathogen Neisseria gonorrhoeae uses homologous recombination for antigenic variation and avoiding immune response. The surface protein pilin (PilE) is regulated by N. gonorrhoeae mismatch repair (MMR) and a G-quadruplex (G4) structure located upstream of the pilE promoter. The study found that ngMutL preferentially binds to pilE G4, which inhibits ngMutL-induced DNA nicking and modulates cleavage positions, suggesting that pilE G4 may regulate the efficiency of pilin antigenic variation.

Summary: Synthetic lethality can be used to target oncogenic drivers in cancer, and in this study, it was found that WRN helicase is required for the survival of MSI CRC cells. Depletion of WRN induces p53 and PUMA, leading to apoptosis in MSI CRC cells. The vulnerability of MSI CRCs to WRN loss is mediated by p53/PUMA-dependent apoptosis. WRN depletion or treatment with ML216 suppresses the growth of MSI CRCs in vitro and in vivo in a p53/PUMA-dependent manner.

Summary: There are currently no therapeutics targeting BKPyV reactivation, and decreasing immunosuppression is the current treatment approach. This study found that the mismatch repair pathway is required for the activation of the DNA damage response during PyV infection. Silencing a component of the mismatch repair pathway resulted in severe DNA damage and reduced the number of infectious viral particles.
