In Escherichia coli and Salmonella enterica, RyeA and RyeB RNAs are encoded on opposite DNA strands at the same locus. We present evidence indicating that the last 23 bp of the ryeB gene, corresponding to an internal portion of the ryeA gene, served repeatedly as the integration site for exogenous DNA during Salmonella evolution and still act as an attachment site for present-day bacteriophages. Interestingly, ryeA sequence and expression are modified upon lysogenization.

However, this damage can be suppressed by methionine sulfoxide reductases MsrA/B, maintaining RecA homeostasis control.ROS not only damage DNA structure directly, but also prevent repair of DNA damage by inhibiting RecA activity.

DNA Breaks-Mediated Fitness Cost Reveals RNase HI as a New Target for Selectively Eliminating Antibiotic-Resistant Bacteria

Summary: The study reveals that the fitness cost of antibiotic resistance mutations in Escherichia coil is strongly correlated with DNA breaks, which are generated via transcription-translation uncoupling and increased formation of RNA-DNA hybrids. Targeting RNase HI enzyme can further increase DNA breaks and the cost of resistance. This research suggests a specific strategy to eliminate resistant bacteria.

Pervasive transcription enhances the accessibility of H-NS-silenced promoters and generates bistability in Salmonella virulence gene expression

Summary: This study analyzed the mechanism of up-regulation of H-NS-silenced Salmonella pathogenicity island 1 (SPI-1) in cells depleted of Rho-cofactor NusG. It was found that when Rho cannot stop the transcription from spurious antisense promoters, it elongates into a H-NS-bound regulatory region of SPI-1, displacing H-NS and leading to the activation of SPI-1 transcription.

Salmonella Type III Secretion Effector SrfJ: A Glucosylceramidase Affecting the Lipidome and the Transcriptome of Mammalian Host Cells

Summary: Type III secretion systems are commonly found in many Gram-negative pathogens and symbionts. Salmonella enterica possess two type III secretion systems associated with virulence, one for host cell invasion and another for intracellular niche maintenance. This study investigates the biochemical function of the SrfJ effector and its impact on mammalian host cells. The results indicate that SrfJ acts as a glucosylceramidase, altering the lipidome and transcriptome of host cells, both in epithelial cells and macrophages during Salmonella infection. Furthermore, analysis of gene expression changes suggests that SrfJ may play a role in protecting Salmonella from host immune defenses.
