Summary: This review summarizes research reports on the epigenetic modifications of spermatozoa during cryopreservation and the potential effects of this process on offspring health. Currently, there are contradictory results regarding the impact of sperm cryopreservation on DNA methylation, and further studies with larger sample sizes are needed to draw definitive conclusions.

Summary: The study evaluated DNA methylation patterns in sperm from men with differential levels of sperm DNA fragmentation index (DFI). The results showed that high DFI in male sperm was associated with significantly altered methylation in specific CpG fragments corresponding to nine genes. This suggests that sperm DNA damage may influence DNA methylation patterns in human sperm.

Summary: This study compared the effects of cryopreservation with dextran and egg yolk on the motility, morphology, and DNA integrity of spermatozoa in dogs of different ages (Chinese Crested breed). The concentration, total number, and motility of fresh spermatozoa decreased, while DNA damage increased in dogs older than 7 years. Cryopreservation using extenders with egg yolk or dextran resulted in a similar decrease in these parameters in the oldest age group. However, considering the standardization of freezing media composition and exclusion of foreign proteins, using dextran for freezing dog spermatozoa is preferable.

Summary: The study revealed that sperm chromatin condensation decreased gradually during in vitro capacitation of ram spermatozoa, while sperm DNA integrity remained intact. Such changes in chromatin condensation appeared faster after sperm cryopreservation.

Summary: This study investigated the effect of seasonal changes on sperm quality and DNA fragmentation dynamics in cats. The results showed that during the breeding season, sperm motility and plasma membrane integrity were higher, while abnormal sperm and head defects were lower. DNA fragmentation was affected by incubation time and reproductive season, suggesting the potential use of this technique in assessing the fertility of feline semen samples for assisted reproduction.

Summary: Oxidative stress is a major cause of male infertility due to its destructive effects on sperm structures and functions. Sperm DNA is highly susceptible to oxidative damage, particularly leading to single- or double-strand DNA breaks, which is a significant issue in male infertility. While methods to detect sperm DNA fragmentation have become important diagnostic tools, there is still no clear consensus on their prognostic value.

Summary: Perinatal smoke/nicotine exposure alters lung development and causes asthma in exposed offspring, transmitted transgenerationally. The mechanism underlying the transgenerational inheritance of perinatal smoke/nicotine-induced asthma remains unknown, but germline epigenetic modulations may play a role.

Summary: This study investigated the potential of mitochondrial antioxidants to protect sperm quality during cryopreservation. The results showed that the addition of BGP-15 or L-carnitine to the cryopreservation medium improved sperm motility and prevented loss of viability. These antioxidants also reduced DNA oxidative damage, with BGP-15 significantly reducing DNA fragmentation. Overall, this study highlights the importance of mitochondrial antioxidants in improving long-term fertility preservation in males.

Summary: Analysis of sperm DNA fragmentation index (DFI) before treatment can provide predictive information on pregnancy rate (PR) and live birth rate (LBR) after intrauterine insemination (IUI). Couples with a baseline DFI below 10 showed significantly higher success rates in IUI.

Summary: Cryopreservation of blood-based DNA can introduce biases in methylation analysis, with a detectable bias towards hypomethylation at the level of individual CpG sites. However, there is no significant change in mean global methylation when analyzed in aggregate. Researchers should be cautious of this hypomethylation bias when profiling methylation in cryopreserved DNA.

Summary: The study on equine spermatozoa revealed that cryopreservation significantly affects redox regulation and oxidative stress in sperm, particularly with significant reductions of Aldo-keto reductase family 1 member B and Superoxide dismutase. This finding supports the importance of controlling redox regulation in improving current cryopreservation procedures.

Summary: This study found that sperm DNA methylation status at specific CpG sites is related to bull fertility and identified seven differently methylated genes in the sperm of subfertile bulls that may lead to altered gene expression and potentially influence embryo development.
