Anti-mullerian hormone and cumulative pregnancy outcome in in-vitro fertilization

Purpose To evaluate the role of Anti-mullerian hormone (AMH) in predicting cumulative pregnancy outcome during in-vitro fertilization (IVF) treatment. Methods Serum AMH levels on day 6 of ovarian stimulation were taken from 180 women undergoing IVF with or without intracytoplasmic sperm injection (ICSI). The main outcome measures were ongoing pregnancy in the fresh cycle, cumulative ongoing pregnancy and ovarian response. Results There was a trend of higher median AMH levels in subjects achieving ongoing pregnancy in the fresh IVF cycle. The median AMH levels were significantly higher in subjects attaining ongoing pregnancy cumulatively and in subjects showing ovarian hyper-response in the stimulated cycle. Areas under the ROC curves were 0.606 and 0.792 for the prediction of cumulative ongoing pregnancy and ovarian hyper-response respectively. Conclusions Serum AMH concentration on day 6 of stimulation was significantly higher in subjects who achieved cumulative ongoing pregnancy in IVF compared to those who did not. Serum AMH is a reasonably good predictor of ovarian hyper-response.

Comparing the intercycle variation of serum anti-Mullerian hormone and antral follicle count measurements over four consecutive menstrual cycles

Summary: The intercycle variation of serum AMH and AFC was compared in this study. The results showed that the repeatability of AMH was significantly better than AFC. Both AMH and AFC had similar predictive performance on poor ovarian response when assessed within three months before IVF treatment.

Identification of risk genes in Chinese nonobstructive azoospermia patients based on whole-exome sequencing

Summary: This study aimed to investigate potential high-risk genes associated with spermatogenesis in idiopathic nonobstructive azoospermia (NOA) patients by whole-exome sequencing. The frequency of cystic fibrosis transmembrane conductance regulator (CFTR) gene pathogenic variant carriers was higher in the NOA patients compared with the healthy controls. In addition, seven potential risk genes that were highly or specifically expressed in the testes were identified, including four previously reported risk genes and three novel risk genes.

Summary: A fast and sensitive HPLC-DMS/MS/MS method was developed for the simultaneous determination of seven androgens and 17-hydroxyprogesterone. Dispersive magnetic solid phase extraction (DMSPE) using Fe3O4@GO nanoparticles and in situ derivatization with Girard's Reagent P were employed. The method showed good precision, accuracy, and extraction recoveries, and was successfully applied to clinical sample analysis.

An online tool for predicting ovarian responses in unselected patients using dynamic inhibin B and basal antimullerian hormone levels

Summary: Mathematical models using early Delta inhibin B and other biomarkers can predict ovarian response in controlled ovarian stimulation. Early Delta inhibin B and basal antimullerian hormone (AMH) levels were the best measures for predicting excessive and poor ovarian responses. These models may help in counseling patients and reducing iatrogenic poor or excessive ovarian responses.

Summary: This study evaluated the association between serum advanced glycation end-products (AGEs) and soluble receptor of AGEs (sRAGE) levels with dysglycaemia and metabolic syndrome in women with polycystic ovary syndrome (PCOS). The results showed that women with dysglycaemia or metabolic syndrome had significantly lower sRAGE levels. However, after adjusting for body mass index, free androgen index and homeostatic model assessment for insulin resistance (HOMA-IR), sRAGE did not have a significant independent association with these outcome measures.

The model showed good predictive ability and could assist clinicians and patients in decision-making and outcome evaluation.

Single-cell whole-genome sequencing, haplotype analysis in prenatal diagnosis of monogenic diseases

Summary: Monogenic inherited diseases are a common cause of congenital disabilities, resulting in significant burdens on affected families. This study demonstrates the feasibility of using cell-based noninvasive prenatal testing (cbNIPT) with single-cell whole-genome sequencing (WGS) and haplotype analysis to diagnose various monogenic diseases prenatally. The results show that WGS performs better than targeted sequencing in terms of genome coverage, allele dropout, and false-positive rates. This research highlights the potential of cbNIPT by WGS and haplotype analysis in the prenatal diagnosis of monogenic diseases.

Summary: Polycystic ovary syndrome (PCOS) is a metabolic disorder affecting fertility. This comprehensive study identified reliable phenotypes that can predict IVF and pregnancy outcomes in PCOS patients. The metabolic syndrome (MetS) phenotype was found to be associated with cumulative live birth, preterm birth, and gestational diabetes mellitus in PCOS patients undergoing IVF (p < 0.05), suggesting its potential for clinical management strategies.

Impact of vaccination against COVID-19 on the outcomes of in vitro fertilization-embryo transfer: a retrospective cohort study

Summary: This study compared the outcomes of IVF-ET and the development of follicles and embryos between vaccinated and unvaccinated groups. The results showed that vaccination against COVID-19 did not have a significant effect on the outcomes of IVF-ET and the development of follicles and embryos. Gender and vaccine type also did not show significant effects.
