Summary: Female infertility is mainly caused by ovulation disorders, with polycystic ovary syndrome (PCOS) being the most prevalent. PCOS is associated with abnormal function of estrogen and estrogen receptors (ERs), affecting cellular activities. Selective estrogen receptor modulators (SERMs) like tamoxifen and clomiphene have shown clinical applications for subfertility with PCOS, but further understanding of the role of ER in PCOS is needed.

Summary: Genetic risk factors for PCOS have phenotypic consequences in men, indicating that they can act independently of ovarian function. Thus, PCOS in women may not always be a primary disorder of the ovaries.

Summary: The study aimed to assess the hormonal and metabolic profile of adolescent girls with PCOS additionally diagnosed with AT, finding higher estradiol and DHEAS concentrations in the AT group. Positive correlation between AT and estradiol concentration was observed, along with a tendency towards higher glucose levels in girls with PCOS and AT, indicating a potential role of estradiol in the development of the autoimmune process. Further research is needed to confirm these findings.

Summary: The MPO G-463A variant is associated with a risk of PCOS in Chinese women, while the CYBA C242T variant is not. The coexistence of the MPO A allele and the CYBA CC genotype is related to an increased risk of PCOS.

Summary: This study aimed to characterize the phenotypic spectrum of PCOS clinical features within and across racial and ethnic groups. Different phenotyping strategies revealed that PCOS symptoms are observed across a severity spectrum that parallels the continuous genetic liability to PCOS in the general population, with racial and ethnic group differences in PCOS symptomology and metabolic health.

Summary: The study found that genetically predicted PCOS is not causally associated with the risk of endometrial cancer in both Europeans and Asians. Subgroup analyses suggest that PCOS may not be linked to the risk of endometrioid endometrial cancer or non-endometrioid endometrial cancer in European ancestry.

Summary: This study successfully established a PCOS model in mice using STZ, with findings indicating that low doses of STZ can induce PCOS features without causing diabetes features. Different doses of STZ treatment had varying effects on body weight, blood glucose levels, and hormone levels in mice.

Summary: The study identified a moderate genetic overlap between PCOS and T2D, and found 11 potential risk variants associated with PCOS, some of which were jointly associated with two phenotypes. These genetic variants are involved in important functions such as lipid metabolism, immune response, and the insulin signaling pathway.

Summary: Polycystic ovary syndrome (PCOS) is a common endocrine disorder in women of reproductive age, affecting fertility and increasing the risk of various health problems. The current understanding of its pathogenesis is still incomplete, leading to difficulties in accurate diagnosis and personalized treatment. This article summarizes the current findings on the genetic, epigenetic, gut microbiota, brain responses, and metabolomics aspects of PCOS, highlighting the challenges in phenotyping and potential treatment approaches. It also discusses the intergenerational transmission of PCOS, providing insights for better management in the future.
