A new highly oxygenated nortriterpenoid, 2-hydroxy-micrandilactone C (1), and three related known triterpenoids (2-4) have been isolated from the leaves and stems of Schisandra chinensis. The structure of the new compound was elucidated by means of NMR and MS spectroscopic analyses.

Sinueretone A, a Diterpenoid with Unprecedented Tricyclo[12.1.0.05,9] pentadecane Carbon Scaffold from the South China Sea Soft Coral Sinularia erecta

Summary: A novel diterpenoid, sinueretone A, was isolated from the South China Sea soft coral Sinularia erecta, along with two new and one known casbane diterpenoids. The structures of the new compounds were determined by extensive spectroscopic analysis and quantum chemical calculations, and a biogenetic relationship between them was proposed. Compounds 1 and 2 showed interesting anti-inflammatory activity in a bioassay.

Summary: A novel norditerpenoid and four polyoxygenated cembranoids were isolated from the Hainan soft coral Sinularia siaesensis, with their structures elucidated through various methodologies. The study also proposed a biosynthetic pathway for the novel compound and demonstrated their inhibitory activity against inflammation in BV-2 microglial cells.

Enhancing the stability of active pharmaceutical ingredients by the cocrystal strategy

Summary: This review discusses the successful application of cocrystallization in addressing the stability issues of active pharmaceutical ingredients (APIs) and the molecular mechanism behind it. By tailored design, delicate modification, and ingenious construction of cocrystals, the stability of APIs can be enhanced. A multidimensional structural understanding is necessary to manipulate interaction strength, molecular conformation, packing styles, and electronic effect. Prominent examples of cocrystals that have successfully dealt with stability problems are summarized, providing insights into vital structural factors in different scenarios and guiding future stable cocrystal design.

Improving the Stability, Dissolution, and Bioavailability of Isotretinoin by Cocrystallization

Summary: This study designed and synthesized four cocrystals of ITT, and found that the cocrystal between ITT and ethyl nicotinate (ENT) showed better chemical stability, dissolution, and absorption. The ITT-ENT cocrystal capsules had higher bioavailability in the fasted state compared to the commercially available ITT capsules.

Stabilizing L-Menthol by Cocrystallization: Supramolecular Cavities Succeeded in Retaining the Volatile Flavor

Summary: L-menthol is an important flavoring additive with diverse applications. The manufacturing and storage of L-menthol face challenges due to its high volatility and low melting point. In this study, six new solid forms of L-menthol were synthesized through cocrystallization, which significantly increased the melting point and reduced the vapor pressure, leading to higher stability under storage conditions. This superior stability can be attributed to the supramolecular cavities assembled by coformers, which restrict the movement of L-menthol molecules through hydrogen bonds.
