Marker-assisted selection of diploid and tetraploid potatoes carrying Rpi-phu1, a major gene for resistance to Phytophthora infestans

The Rpi-phu1 gene originates from an interspecific hybrid between Solanum stenotomum and S. phureja, and confers a high level of resistance to Phytophthora infestans (late blight) in potato. The Rpi-phu1 was introduced by crossing at the diploid level into the S. tuberosum gene pool and then transferred to the tetraploid level by means of 2n gametes. Tetraploid lines carrying the Rpi-phu1 were selected for further crosses. A molecular marker GP94, linked in mapping population 97-30 with the Rpi-phu1 (6.4 cM), was applied to other unselected populations (2 diploid and 1 tetraploid), and was shown to be useful in marker-assisted selection (MA S) of the resistant individuals. GP94 was applied also in commercial breeding in 2 tetraploid populations. Although the marker allele of GP94, characteristic for the Rpi-phu1 presence, was rare in the S. tuberosum gene pool, it seemed to be common in 2 other sources of resistance to late blight: S. ruiz-ceballosii and S. kurtzianum.

Identification and pathogenicity of Fusarium spp. associated with tuber dry rot and wilt of potato in Algeria

Summary: This study identified and characterized the main pathogens causing potato diseases in Algeria, with Fusarium sambucinum being the most common species. Fusarium oxysporum and F. nygamai were the most aggressive in causing potato wilt, while F. sambucinum was the most aggressive in causing potato tuber dry rot. This research provides valuable insights into the management of potato diseases in Algeria.

Reference gene selection for miRNA and mRNA normalization in potato in response to potato virus Y

Summary: This study evaluated candidate reference genes for quantifying the expression profiles of coding and non-coding genes in potato responding to potato virus Y inoculation. The most stable reference genes were determined to be sEF1a, sTUBb and seIF5, while the least stable ones were sPP2A, sSUI1 and sGAPDH. Using the same reference gene allowed for normalization of both miRNA and mRNA levels from a single RNA sample.

QTLs for potato tuber resistance to Dickeya solani are located on chromosomes II and IV

Summary: This research identified QTLs for potato tuber resistance to D. solani and for tuber starch content, as well as investigated the relationship between these traits. Prominent QTLs for disease severity and incidence were identified on potato chromosome IV, with additional QTLs for starch content on other chromosomes. The findings suggest potential for marker development and further study in potato breeding programs.

Over 50 Years of Potato Parental Line Breeding Programme at the Plant Breeding and Acclimatization Institute in Poland

Late blight resistance genes in potato breeding

Summary: Using resistance genes targeting conservative effectors of Phytophthora infestans and constructing gene pyramids can provide durable, broad-spectrum resistance. Genetic engineering techniques enable easier transfer of resistance genes to cultivated varieties, increasing the durability and spectrum of potato resistance to evolving P. infestans strains.

Summary: Wild potato relatives are potential sources of desirable agricultural traits for potato breeding. We described 29 seed accessions of 26 Solanum species from the VIR potato collection and preserved in Poland. The evaluation included resistance to disease, tuber flesh color, enzymatic discoloration, and tuber dormancy. We found valuable potato relatives with resistance, white flesh, and longer dormancy periods.

Polish Cryobanks: Research and Conservation of Plant Genetic Resources

Summary: The use of liquid nitrogen (LN) in plant gene banks is currently limited, with only 20 out of 500 banks in Europe utilizing cryogenic temperatures. Poland has gradually developed its gene banking system since the 1990s, mainly focusing on seed collection. This review provides the first summary of gene cryobank activities and resources in Poland, highlighting the use of LN in plant protection programs and presenting the characteristics of four existing gene cryobanks in the country.

Population Structure of Phytophthora infestans from a Single Location in Poland Over a Long Period of Time in Context of Weather Conditions

Summary: This study investigated 237 isolates of Phytophthora infestans collected from an unprotected experimental field in Poland over 15 growing seasons. The results showed 89 unique genotypes, with 87 isolates belonging to genotype 34_A1. The study also analyzed the variability in metalaxyl resistance and virulence among isolates of the P. infestans genotype 34_A1.
