Cancer stem cells: the theory and perspectives in cancer therapy

The cancer stem cell theory elucidates not only the issue of tumour initiation and development, tumour's ability to metastasise and reoccur, but also the ineffectiveness of conventional cancer therapy. This review examines stem cell properties, such as self-renewal, heterogeneity, and resistance to apoptosis. The 'niche' hypothesis is presented, and mechanisms of division, differentiation, self-renewal and signalling pathway regulation are explained. Epigenetic alterations and mutations of genes responsible for signal transmission may promote the formation of cancer stem cells. We also present the history of development of the cancer stem cell theory and discuss the experiments that led to the discovery and confirmation of the existence of cancer stem cells. Potential clinical applications are also considered, including therapeutic models aimed at selective elimination of cancer stem cells or induction of their proper differentiation.

Summary: The aim of this study was to determine the suitability of the aCGH technique for prenatal diagnosis and assess the frequency of chromosomal aberrations not included in the diagnostic report. The study included 7400 prenatal cases and found a 27.2% chromosomal aberration detection rate, with 71.2% being numerical aberrations and 28.8% being structural aberrations. Clinically significant CNVs were detected in 6.8% of cases, and variants of unknown clinical significance were found in 0.7% of cases. The detection rate of clinically significant submicroscopic CNVs varied among different indications for invasive testing.

Genetic data protection as an indispensable element of genomic medicine development

Summary: The progress in molecular biology has changed our understanding of human diseases. Whole genome analyses have allowed for the identification of unknown diseases. Molecular diagnosis is replacing clinical diagnosis in difficult cases. Construction and sharing of large databases are crucial for genetic research and clinical characteristics of rare diseases. Legal and ethical issues surrounding genetic testing and personal data protection must be addressed. This paper explores the benefits and risks of international sharing of genetic databases and discusses legal provisions in the EU, US, and China.
