Summary: This study describes an unusual biotype of KPC-2-producing Klebsiella pneumoniae isolated in Corrientes, Argentina. The isolates belong to the high-risk hospital-associated clonal lineage ST340/CC258. Genomic characterization revealed multiple acquired resistance genes and mutations, as well as the presence of virulence determinants. Additionally, an unusual plasmid type was identified.

Summary: In this study, we evaluated phenotypic methods for detecting coexpression of KPC-2 and NDM-1 in members of the K. pneumoniae complex. We found that inhibition by AVI, DPA, and EDTA may provide simple and inexpensive methods for the presumptive detection of coexpression of KPC-2 and NDM-1 in members of the K. pneumoniae complex.

Summary: Designing novel beta-lactamase inhibitors (BLIs) is an accepted strategy to combat the resistance of Gram-negative bacteria to cephalosporin and carbapenem. This study evaluates the activity of two alpha-amido-beta-triazolylethaneboronic acid transition state inhibitors (S02030 and MB_076) against KPC-2 and CTX-M-96 beta-lactamases. The inhibitors show potent activity and their structures provide insights into the design of effective BLIs.

Summary: Antimicrobial resistance is a significant global health threat. Analyzing individual antibiotic resistance genes (ARGs) is crucial for combating this threat. By analyzing ARGs in 757 sewage samples from various locations, we found regional differences in resistomes that are influenced by drug classes and taxonomic variations.

Summary: This study describes an outbreak of ceftazidime-avibactam-resistant KPC variants of Klebsiella pneumoniae in a hospital in Argentina. The detection of these bacteria is challenging for the laboratory, and the history of previous infections and resistance testing may help raise awareness of this possibility.

Biochemical and Structural Characterization of CRH-1, a Carbapenemase from Chromobacterium haemolyticum Related to KPC β-Lactamases

Summary: This study analyzed the kinetic behavior and crystallographic structure of a class A CRH-1 beta-lactamase. The results showed that CRH-1 can effectively hydrolyze penicillins, cephalosporins, and carbapenems, similar to KPC-2. However, CRH-1 is poorly inhibited by clavulanic acid and efficiently inhibited by avibactam. The findings highlight the biochemical and structural similarities of CRH-1 and KPC-2 and the potential clinical impact of this carbapenemase.

Summary: Carbapenemase resistance in Enterobacterales is a global public health problem. This study evaluated a protocol for detecting Klebsiella pneumoniae carbapenemase (KPC) using MALDI-TOF MS technology. The KPC direct detection showed a sensitivity and specificity of 100% from patients' positive blood cultures, short-term cultures, and colonies. This study is significant in the context of multi-resistance and highlights the need for actions to reduce the spread of antibiotic resistance.

Summary: This study characterized isogenic strains of S. aureus with vancomycin resistance, both in vivo and in vitro, and found genetic changes associated with cell wall and phosphate metabolism, staphylococcal growth, and biofilm formation. These changes are not commonly associated with vancomycin resistance in S. aureus.

Diversity of genetic platforms harboring the blaPER-2 gene in Enterobacterales and insights into the role of ISPa12 in its mobilization and dissemination

Summary: The production of PER-like extended-spectrum β-lactamases has been linked to reduced susceptibility to certain drugs. The blaPER-2 gene has been mainly found in Argentina and neighboring countries but little is known about its transmission through different plasmid groups. A study analyzed the genetic platforms of PER-producing bacteria and found that the blaPER-2 gene is encoded on plasmids of different incompatibility groups, indicating potential dissemination through various plasmids. The study also identified the role of certain insertion sequence elements in the mobilization and further dissemination of the blaPER-2 gene.

Summary: Two novel variants of KPC, KPC-113 and KPC-114, associated with resistance to CZA, were identified in clinical isolates of Klebsiella pneumoniae in Brazil. While both variants showed resistance to several antibiotics, KPC-114 remained susceptible to imipenem and meropenem.
