Prevalence of blaKPC-2, blaKPC-3 and blaKPC-30-Carrying Plasmids in Klebsiella pneumoniae Isolated in a Brazilian Hospital

Summary: This study reports the genomic context of KPC coding genes bla(KPC-2), bla(KPC-3) and bla(KPC-30) in multidrug-resistant Klebsiella pneumoniae isolates from Brazil, with a focus on the prevalence and dissemination of bla(KPC)-harboring plasmids. It demonstrates the risk of horizontal spread of KPC coding genes and the high synteny of plasmid backbones among isolates, including the recent spread of bla(KPC-3) and bla(KPC-30).

Silent circulation of BKC-1-producing Klebsiella pneumoniae ST442: molecular and clinical characterization of an early and unreported outbreak

Willames M. B. S. Martins, Michael H. Lenzi, Ana C. Narciso, Priscila P. Dantas, Diego O. Andrey, Qiu E. Yang, Kirsty Sands, Eduardo A. Medeiros, Timothy R. Walsh, Ana C. Gales

Summary: This study describes the undetected outbreak of a rare carbapenemase BKC-1-producing Klebsiella pneumoniae ST442 clone, occurring in a Brazilian hospital. The misidentification of BKC-1 may have contributed to the spread of resistant clones, emphasizing the importance of correct and rapid identification of antimicrobial resistance mechanisms in hospitals.

Sequencing of fosA: A Rapid and Inexpensive Method for Discriminating Klebsiella pneumoniae CC258 from Other Clones

Aghata Cardoso da Silva Ribeiro, Fernanda Fernandes Santos, Ikechukwu Benjamin Moses, Luciene Andrade da Rocha Minarini, Ana Cristina Gales

Summary: In this study, a new fast, economic, and effective typing technique for carbapenem-resistant Klebsiella pneumoniae (CR-KPN) was explored by analyzing the fosA(KP) sequence. The results showed that chromosomal fosA(KP) sequences are highly conserved in different STs and CCs, providing a useful tool to identify the epidemiological STs for KPN and discriminate them from CC258/11.

In Vitro and In Vivo Synergism of Fosfomycin in Combination with Meropenem or Polymyxin B against KPC-2-Producing Klebsiella pneumoniae Clinical Isolates

Aghata Cardoso da Silva Ribeiro, Yohanna Carvalho dos Santos Aoun Chikhani, Tiago Barcelos Valiatti, Andre Valencio, Mariana Neri Lucas Kurihara, Fernanda Fernandes Santos, Luciene Andrade da Rocha Minarini, Ana Cristina Gales

Summary: In this study, the activity of fosfomycin in combination with meropenem or polymyxin B against KPC-KPN was evaluated. The results showed that the combination of fosfomycin and meropenem restored meropenem activity and exhibited a synergistic effect with fosfomycin and polymyxin B. In the Galleria mellonella model, the combination of fosfomycin and meropenem had a higher survival rate for larvae, while fosfomycin and polymyxin B combination had a more significant biofilm disruption effect.

Summary: Isavuconazole is the only FDA-approved antifungal for treating invasive mucormycosis. A global collection of Mucorales isolates was evaluated, and it was found that Isavuconazole displayed good inhibitory activity against most isolates, although there was variation among different species.
