Summary: Infection of the female genital tract by Chlamydia trachomatis can lead to severe fibrotic sequelae, and it remains unclear if intrinsic properties of the upper genital tract exacerbate the fibrotic response. This study found that cervical epithelial cells have enhanced expression of fibrosis-associated signal factors and infection-mediated induction of fibrosis-related genes. The activation of YAP, a transcriptional cofactor, by chlamydial infection in cervical epithelial cells was identified as a potential driver of fibrotic gene expression. These findings provide new insights into the mechanism of Chlamydia-induced fibrosis and suggest YAP as a potential therapeutic target for preventing scarring of the female genital tract.

Summary: Adhesion and colonization of host cells by pathogenic bacteria rely on protein-protein interactions (PPIs). This study developed a filtering process to obtain a reliable list of domains and motifs participating in PPIs between bacteria and human cells. Structural analysis revealed that human proteins involved in these interactions are rich in alpha helix and disordered regions. The most significant domain-domain interactions were validated, but verifying domain-motif interactions was more challenging. Post-translational modifications, especially phosphorylation, also play a role in these interactions.

To address the limited knowledge of host-pathogen interactomes, we developed HPIPred, a numerical encoding-based host-pathogen PPI prediction tool. Through screening the entire proteomes of Homo sapiens and Pseudomonas aeruginosa PAO1, we generated a highly connected network topology of host-pathogen interactome using HPIPred.

A peptide-based quorum sensing inhibitor was designed, which attenuated Bc infectivity in mouse models by suppressing virulence factor expression. These findings reveal a novel strategy to limit Bc virulence and enhance host defense.
