On November 8-9, 2007, the Society for Acupuncture Research (SAR) hosted an international conference to mark the tenth anniversary of the landmark NIH [National Institutes of Health] Consensus Development Conference on Acupuncture. More than 300 acupuncture researchers, practitioners, students, funding agency personnel, and health policy analysts from 20 countries attended the SAR meeting held at the University of Maryland School of Medicine, Baltimore, MD. This paper summarizes important invited lectures in the area of basic and translational acupuncture research. Specific areas include the scientific assessment of acupuncture points and meridians, the neural mechanisms of cardiovascular regulation by acupuncture, mechanisms for electroacupuncture applied to persistent inflammation and pain, basic and translational research on acupuncture in gynecologic applications, the application of functional neuroimaging to acupuncture research with specific application to carpal-tunnel syndrome and fibromyalgia, and the association of the connective tissue system to acupuncture research. In summary, mechanistic models for acupuncture effects that have been investigated experimentally have focused on the effects of acupuncture needle stimulation on the nervous system, muscles, and connective tissue. These mechanistic models are not mutually exclusive. Iterative testing, expanding, and perhaps merging of such models will potentially lead to an incremental understanding of the effects of manual and electrical stimulation of acupuncture needles that is solidly rooted in physiology.

Prenatal androgen exposure causes a sexually dimorphic transgenerational increase in offspring susceptibility to anxiety disorders

Sanjiv Risal, Maria Manti, Haojiang Lu, Romina Fornes, Henrik Larsson, Anna Benrick, Qiaolin Deng, Carolyn E. Cesta, Mina A. Rosenqvist, Elisabet Stener-Victorin

Summary: This study found that daughters of women with PCOS have an increased risk of anxiety disorders, possibly transmitted transgenerationally through the female germline. Using a PCOS mouse model, researchers observed anxiety-like behavior in female offspring in the androgenized lineage and male offspring in both the androgenized and obese lineages. Additionally, dysregulated genes related to anxiety-like behavior were identified in the amygdala of female offspring in the androgenized lineage and male offspring in the obese lineage.

Maternal adiponectin prevents visceral adiposity and adipocyte hypertrophy in prenatal androgenized female mice

Yanling Wu, Belen Chanclon, Peter Micallef, Elisabet Stener-Victorin, Ingrid Wernstedt Asterholm, Anna Benrick

Summary: The study demonstrates that elevated levels of adiponectin in utero can improve insulin sensitivity, reduce body weight and fat mass gain in adult offspring, and protect against PNA-induced visceral adiposity in the offspring.

Is there a shared genetic basis and causal relationship between polycystic ovary syndrome and psychiatric disorders: evidence from a comprehensive genetic analysis

X. Jiang, Q. Deng, E. Stener-Victorin

Summary: This study examined the genetic basis and causal relationship between polycystic ovary syndrome (PCOS) and a range of psychiatric disorders, finding no significant genetic correlation or causal relationship between them, despite some associations in certain contexts. While PCOS seems to be related to depression genetically, this effect may be mediated by BMI.

Epigenetic inheritance of polycystic ovary syndrome - challenges and opportunities for treatment

Elisabet Stener-Victorin, Qiaolin Deng

Summary: Polycystic ovary syndrome (PCOS) is a major cause of female infertility worldwide, with high prevalence and economic burden. The etiology of PCOS involves genetic and developmental factors, with hyperandrogenism playing a key role. Current treatments are limited, highlighting the need for more effective preventative and targeted therapies.

Transmission of Polycystic Ovary Syndrome via Epigenetic Inheritance

Elisabet Stener-Victorin, Qiaolin Deng

Summary: A recent study has found that late gestation exposure to anti-Mullerian hormone can result in the transgenerational transmission of a PCOS-like phenotype in mice, with altered DNA methylation being the underlying mechanism. Epigenetics-based therapy has shown promise in reversing some PCOS-like traits in F-3 female mice.

Adiponectin Deficiency Alters Placenta Function but Does Not Affect Fetal Growth in Mice

Man Mohan Shrestha, Sanne Wermelin, Elisabet Stener-Victorin, Ingrid Wernstedt Asterholm, Anna Benrick

Summary: Adiponectin deficiency affects fetal growth and placental function, particularly in obese individuals. Low levels of adiponectin lead to placental dysfunction and increased fetal triglyceride load, which could have long-term effects on the offspring's metabolism.

A genome-wide cross-trait analysis identifies shared loci and causal relationships of type 2 diabetes and glycaemic traits with polycystic ovary syndrome

Qianwen Liu, Bowen Tang, Zhaozhong Zhu, Peter Kraft, Qiaolin Deng, Elisabet Stener-Victorin, Xia Jiang

Summary: The genetic link between type 2 diabetes, glycaemic traits and PCOS was found, influenced by both biological pleiotropy and causal mediation, some of which is independent of BMI. The study highlights the importance of controlling fasting insulin levels to mitigate the risk of PCOS, as well as screening for and monitoring type 2 diabetes in all women with PCOS, irrespective of BMI.

Transcriptomic survey of key reproductive and metabolic tissues in mouse models of polycystic ovary syndrome

Yu Pei, Sanjiv Risal, Hong Jiang, Haojiang Lu, Eva Lindgren, Elisabet Stener-Victorin, Qiaolin Deng

Summary: Excessive androgen production and obesity play a key role in the development of polycystic ovary syndrome (PCOS). Various mouse models, including prenatal androgenized, peripubertal androgenized, overexpression of nerve growth factor in theca cells, and diet-induced maternal obesity, were used to study PCOS. Transcriptome analysis revealed differentially expressed genes in the hypothalamus, adipose tissue, ovary, and oocytes, with the ovaries and adipose tissues showing the most significant changes. The Ms4a6e gene, found in all mouse models and in women with PCOS, suggests a conserved disease function in PCOS.

Circulating triglycerides are associated with human adipose tissue DNA methylation of genes linked to metabolic disease

Tina Ronn, Alexander Perfilyev, Josefine Jonsson, Karl-Fredrik Eriksson, Sine W. Jorgensen, Charlotte Brons, Linn Gillberg, Allan Vaag, Elisabet Stener-Victorin, Charlotte Ling

Summary: This study found an association between DNA methylation levels in adipose tissue and circulating lipid levels, and identified novel epigenetic changes associated with lipid levels. These findings provide insights into the relationship between dysregulation of circulating lipids and epigenetic mechanisms in adipose tissue, and have important implications for the study of metabolic syndrome.

Body image concerns in women with polycystic ovary syndrome: a systematic review and meta-analysis

Summary: This systematic review and meta-analysis compared body image concerns among women with and without polycystic ovary syndrome (PCOS). The results showed that women with PCOS reported higher dissatisfaction with appearance evaluation and appearance orientation. They also had higher dissatisfaction with overweight preoccupation, lower body area satisfaction, and body weight classification compared to women without PCOS. These findings highlight the importance of addressing body image concerns in the clinical care of PCOS.
