Esterification Enhanced Intestinal Absorption of Ginsenoside Rh2 in Caco-2 Cells without Impacts on Its Protective Effects against H2O2-Induced Cell Injury in Human Umbilical Vein Endothelial Cells (HUVECs)

Ginsenoside Rh2 and its octyl ester derivative (Rh2-O) were investigated for their transcellular transport in the Caco-2 cell system and their protective effect against oxidative stress in human umbilical vein endothelial cells (HUVECs). Results showed that the transport rates for apical-to-basolateral (AP-BL) flux of Rh2 (0.21 x 10(-6) cm/s) was enhanced by the synthesis of its esterified derivative Rh2-O (1.93 x 10-6 cm/s) over the concentrations of 10-50 mu M. In addition, both Rh2 and its esterified derivative Rh2-O exhibited similar protective effects against oxidative damage induced by H2O2. Pretreatment of Rh2 and Rh2-O significantly decreased the activation of caspase-3 known to play a key role in H2O2-induced cell apoptosis. These results were consistent with that of a flow cytometry assay analyzing HUVECs apoptosis. The present study demonstrated that the absorption of ginsenoside Rh2 in vitro can be significantly enhanced by synthesis of its ester derivative. Meanwhile, no significant discrepancy between Rh2 and Rh2-O on their bioactivities against the oxidative damage induced by H2O2 was observed, which means that esterification of Rh2 might have a higher bioavailability than Rh2 in vitro without impacts on pharmaceutical actions.

Red Ginseng Improves D-galactose-Induced Premature Ovarian Failure in Mice Based on Network Pharmacology

Summary: This study evaluated the ameliorative effect and molecular mechanism of red ginseng extract on D-galactose-induced premature ovarian failure using network pharmacology analysis. Ginsenosides, sugar, and amino acid derivatives in red ginseng were identified as important active ingredients. The study constructed a protein-protein interaction network and identified core targets related to premature ovarian failure. In vivo experiments confirmed that red ginseng extract can increase estradiol levels and reduce follicle-stimulating hormone levels by reducing oxidative stress, apoptosis, and senescence signaling pathways.

Review on potential effects of traditional Chinese medicine on glaucoma

Platycodin D stimulates AMPK activity to inhibit the neurodegeneration caused by reactive oxygen species-induced inflammation and apoptosis

Summary: This study investigated the regulatory effects of Platycodin D (PD) on neurodegeneration caused by reactive oxidative stress (ROS). In vivo and in vitro experiments demonstrated that PD could improve memory, reduce cell apoptosis and ROS production, and inhibit inflammation. PD enhanced the antioxidant ability by activating AMPK. This study suggests that PD may be a potential pharmaceutical agent to treat ROS-induced neurodegeneration.

Lobetyolin, a Q-marker isolated from Radix Platycodi, exerts protective effects on cisplatin-induced cytotoxicity in HEK293 cells

Summary: This study investigated the protective effect of lobetyolin (LBT) against cisplatin-induced cytotoxicity in HEK293 cells. Results showed that LBT improved the viability of HEK293 cells, decreased MDA levels, decreased GSH content, and suppressed ROS levels caused by cisplatin. Molecular docking analysis revealed a strong binding affinity between LBT and the NF-κB protein. LBT also inhibited inflammation and apoptosis in HEK293 cells.

Amelioration of Cisplatin-Induced kidney injury by Arabinogalactan based on network pharmacology and molecular docking

Summary: This study investigated the potential protective mechanism of arabinogalactan (AG) on acute kidney injury (AKI) induced by cisplatin in mice. The results showed that pretreatment with AG could reduce kidney injury and oxidative stress induced by cisplatin, as well as restore the expression of anti-apoptotic proteins and decrease the expression of pro-apoptotic proteins in renal tissues.

Identification of OLA1 as a Novel Protein Target of Vitexin to Ameliorate Dextran Sulfate Sodium-Induced Colitis with Tissue Thermal Proteome Profiling

Summary: This study revealed the anti-inflammatory properties of vitexin in a mouse model of colitis and its ability to enhance the expression of barrier-associated proteins. Through thermal proteome profiling and molecular docking, OLA1 was identified as a potential protein target for vitexin. Further experiments demonstrated that OLA1 can increase Nrf2 protein expression, thereby mediating the anti-inflammatory effects of vitexin.

Structural Characterization of Zinc-Sucrose Complex and Its Ability to Promote Zinc Absorption in Caco-2 Monolayer Cells and Mice

Summary: Sucrose is shown to be a stable metal-ion chelating agent that can enhance the absorption of metal ions. This study explores the structure of the zinc-sucrose complex and its ability to promote zinc absorption in cells and mice. The complex increases zinc uptake and transport, upregulates genes and proteins involved in zinc absorption, and interacts with zinc transport proteins. These findings offer valuable insights into the mechanisms of zinc absorption and suggest potential applications in treating zinc deficiency.

Human Milk Oligosaccharides: A Critical Review on Structure, Preparation, Their Potential as a Food Bioactive Component, and Future Perspectives

This article compares oligosaccharides in human and other mammalian milk, summarizes the synthetic strategies for accessing HMOs, reviews the potential use and molecular mechanisms of HMOs in different diseases, and discusses the current challenges and prospects of HMOs in basic research and application.

Effects of Human Milk Fat Substitutes on Lipid Metabolism in First-Weaned Rats

Summary: Although formula-fed infants are more prone to obesity than breastfed ones, this study found that administering human milk fat substitutes (HMFS) did not affect body weight in rats, but significantly decreased body fat content. The study also revealed the potential mechanism by which HMFS affects body fat buildup and promotes lipid metabolism in the liver. These findings underscore the nutritional benefits of HMFS for infants.

Platycodin D inhibits HFD/STZ-induced diabetic nephropathy via inflammatory and apoptotic signaling pathways in C57BL/6 mice

Summary: This study aimed to investigate the improvement effects and regulatory mechanisms of Platycodin D (PD) on kidney injury in diabetic nephropathy. The results showed that PD can lower blood glucose and insulin resistance levels, improve lipid levels and renal function, and inhibit the development of diabetic nephropathy by regulating NF-cB and apoptotic signaling pathways.

1-Oleate-2-palmitate-3-linoleate glycerol improves lipid metabolism and gut microbiota and decreases the level of pro-inflammatory cytokines

Summary: The most abundant triacylglycerol (TAG) in Chinese human milk is 1-oleate-2-palmitate-3-linoleate (OPL), which is different from other countries where 1,3-oleate-2-palmitate (OPO) is the most abundant TAG. This study investigated the effects of an OPL-supplemented diet on mice's nutritional outcomes and found that high OPL (HOPL) diet improved lipid metabolism and gut bacteria, reducing the level of pro-inflammatory cytokines. Furthermore, the study revealed a relationship between gut bacteria, lipidome profile, and nutritional outcomes.

Differences in Absorption and Metabolism between Structured 1,3-Oleate-2-palmitate Glycerol and 1-Oleate-2-palmitate-3-linoleate Glycerol on C57BL/6J Mice

Lipidome analysis revealed changes in glycerophospholipid levels in the liver of HMFS-fed rats. Furthermore, HMFS administration promoted lipid metabolism in the rat liver at both the gene and protein levels.
