Summary: This study aimed to investigate the role of white matter (WM) structure and function in the neurobiological mechanisms of major depressive disorder (MDD). Results showed abnormal amplitude of low frequency fluctuation (ALFF) in specific frequency bands in WM of drug-naive first-episode MDD patients. It was also found that functional damage of WM may precede structural damage in MDD, and ALFF value in superior longitudinal fasciculus (SLF) was negatively correlated with HAMA-2 score.

Summary: In this study, a negative correlation between abnormal FC in the DMN and severity of depression symptoms in treatment-naive MDD patients was found. The FC in the left MOFG and right SMG was decreased in depressive patients compared to controls, with a negative correlation between abnormal FC in the SMG and HAM-D scores. No increase in FC of the DMN was observed in treatment-naive, first-episode MDD patients. This study sheds light on the relevance of DMN in understanding the pathophysiology of cognitive impairment in MDD.
